摘  要:本研究旨在分析多胎萨福克母羔初情期启动中生殖激素的水平变化,及与BMPR-IB基因突变的相关性,为阐明绵羊初情期启动的遗传机制提供基础。试验应用PCR-RFLP法对461只3.5月龄半舍饲多胎萨福克羔羊进行BMPR-IB基因A746G位点分型,选择体重相近的29只母羔,应用公羊试情法确定初情期,酶联免疫吸附法(ELISA)测定初情前1d、初情期、初情后1d和初情后2d相同时间点及不同基因型母羔外周血中促性腺激素释放激素(GnRH)、促卵泡素(FSH)、黄体生成素(LH)、孕酮(P4)、雌二醇(E2)和催乳素(PRL)浓度,统计分析羔羊初情前后四个时间点外周血中生殖激素含量变化,及BMPRIB三种基因型间生殖激素含量的差异。结果表明:在多胎萨福克母羔外周血中,初情前1d的GnRH、P4和PRL含量高,初情期FSH、LH和E2含量高。BMPR-IB基因BB基因型FSH、P4和E2含量高,B+基因型GnRH、PRL含量高。但初情期前后四个时间点及三种基因型间的六种生殖激素含量均差异不显著(P>0.05)。说明BMPR-IB基因突变未引起初情期母羔生殖激素含量的显著变化,母羔初情期前后的外周血生殖激素水平没有发生显著变化,与初情期母羔发情表现不明显有关。The purpose of this paper is to provide the basis for puberty onset in sheep by analyzing the changes of serum reproductive hormones secretion and its relationship with BMPR-IB gene mutation at puberty in lambs.The genotypes of BMPR-IB gene were detected in 4613.5-month-old prolific Suffolk lambs in semi-barn-fed by PCR-RFLP method.The pubertal time of 29 female lambs with similar weight in the study was determined by detection estrus of ram.The concentrations of GnRH,FSH,LH,P4,E2 and PRL in serum were measured by ELISA method among 4 time points of puberty and BMPR-IB genotypes,the concentration differences of reproductive hormone were analyzed among four time point of prepuberty,puberty and metestrus,and among different genotypes of BMPR-IB gene at the same time point.The results showed as follows:there were high contents of GnRH,P4 and PRL in prepuberty,and high contents of FSH,LH and E2 in puberty in the peripheral blood of female lambs.Three genotypes of BMPR-IB gene were detected in prolific Suffolk sheep.Contents of FSH,P4 and E2 were high in BB genotype,contents of GnRH and PRL were high in B+genotype.Contents of six reproductive hormones among 4 time points before and after puberty,and among three genotypes there were no significant difference(P>0.05).It signified that the mutation of BMPR-IB gene did not significantly cause the content changes of reproductive hormones during the initiation of puberty in lambs.And there were no significant changes in level of reproductive hormones inperipheral blood during the puberty onset of prolific Suffolk lambs,which were related to unobvious estrous symptoms at puberty in prolific Suffolk female lambs.
