摘 要:以红花国槐‘姹紫1号’(Sophora japonica‘chazi 1’)组培苗为试验材料,以6-苄氨基嘌呤(6-BA)、萘乙酸(NAA)、吲哚丁酸(IBA)等植物生长调节剂,采用单因素试验、响应面分析法,分析不同质量浓度组合的植物生长调节剂对‘姹紫1号’增殖培养基的不定芽增殖率、组培苗苗高增长量的影响,遴选红花国槐‘姹紫1号’组培快繁体系。结果表明:6-苄氨基嘌呤质量浓度对不定芽增殖率影响最为显著,萘乙酸与吲哚丁酸的交互作用对不定芽增殖率影响显著;吲哚丁酸质量浓度对苗高增长量的影响最为显著,6-苄氨基嘌呤与萘乙酸的交互作用、萘乙酸与吲哚丁酸的交互作用对苗高增长量的影响显著。增殖最优植物生长调节剂组合为6-苄氨基嘌呤质量浓度1.96mg/L、萘乙酸质量浓度0.45mg/L、吲哚丁酸质量浓度0.54mg/L;红花国槐‘姹紫1号’增殖培养基的不定芽增殖率为420.31%,苗高增长量为6.08 mm。实际测量值与模型预测值基本符合,应用响应面法优化红花国槐‘姹紫1号’增殖培养基切实可行。In order to select the optimal tissue culture and rapid propagation system of Sophora japonica cv.‘Chazi 1’,the tissue culture seedlings of S.japonica were used as experimental materials,supplemented with 6-benzylaminopurine(6-BA),naphthalene acetic acid(NAA),indole butyric acid(IBA)and other plant growth regulators to analyze the effects of different concentrations of plant growth regulators on the proliferation rate of adventitious buds and the height growth of tissue culture seedlings of‘Chazi 1’by single factor test and response surface analysis.The concentration of 6-benzylaminopurine had the most significant effect on the proliferation rate,and the interaction between naphthalene acetic acid and indole butyric acid had a significant effect on the proliferation rate.Indole butyric acid concentration had the most significant effect on seedling height growth.The interaction between 6-benzylaminopurine and naphthalene acetic acid and the interaction between naphthalene acetic acid and IBA had significant effect on seedling height growth.The optimal plant growth regulator formula for proliferation was 6-benzylaminopurine concentration 1.96 mg/L,naphthalene acetic acid concentration 0.45 mg/L and indole butyric acid concentration 0.54 mg/L.Under this condition,the proliferation rate of S.japonica proliferation medium was 420.31%,and the growth of seedling height was 6.08 mm.The actual measured value is basically consistent with the predicted value of the model.It is feasible to optimize the proliferation medium of S.japonica by response surface methodology.
