摘 要:观赏海棠种质资源丰富,表型鉴定与分类难度大。本试验以74个观赏海棠品种为材料,利用SSR分子标记以及表型性状测定,分析观赏海棠的遗传多样性并构建指纹图谱。利用20对谱带清晰、多态性好的引物对74个种(品种)进行PCR扩增及遗传多样性分析,共检测到766个等位基因,扩增出的多态位点百分率达100%,每个位点的平均等位基因数为38.3个。遗传多样性信息含量变化幅度为0.86~0.97,基因多样性指数平均值为0.94,表现出丰富的遗传多样性。从分子水平上探讨其亲缘关系,基于SSR的聚类结果可将74个观赏海棠品种分为四大类。最终利用3对SSR引物成功地构建了74个观赏海棠的指纹图谱,为海棠种质资源的保护、开发利用及创新提供参考和依据。It is difficult to identify and classify from phenotype as ornamental crabapple is rich in germplasm resources.Genetic diversity of 74 varieties was analysed using SSR molecular markers and phenotypic traits,and their fingerprints were constructed.PCR amplification and genetic diversity analysis of 74 species(variety)was carried out with 20 pairs of primers of clear bands and good polymorphism.A total of 766 alleles were detected,and the percentage of amplified polymorphic loci reached 100%,the average number of alleles per locus was 38.3.The variation range of polymorphism information content was 0.86-0.97.The average value of gene diversity was 0.94,which showed abundant genetic diversity.Based on SSR clustering results by exploring their kinships at the molecular level,74 ornamental crabapple varieties coud be divided into four categories.The fingerprints of 74 orna mental crabapple varities were successfully constructed using 3 pairs of SSR primers.It would provide reference and basis for the protection,development,utilization and innovation of ornamental crabapple germplasm resources.
