4种中药单体对抗原提呈细胞免疫激活作用的研究

摘  要:目的在细胞水平上评估白杨素、蔓荆子黄素、刺芒柄花素和北美圣草素4种中药单体对抗原提呈细胞(APCs)的免疫激活能力,从中筛选出具有免疫激活作用的中药单体。方法将RAW264.7细胞和DC2.4细胞按照处理方式的不同,分为白杨素组、蔓荆子黄素组、刺芒柄花素组和北美圣草素组,再根据各中药单体的浓度不同分为低、中、高三个亚组,同时设置阴性对照亚组和脂多糖阳性对照亚组,处理48 h后,分别通过流式细胞仪检测各亚组RAW264.7细胞和DC2.4细胞表面CD80、CD86、主要组织相容性复合体Ⅰ(MHCⅠ)和MHCⅡ分子的表达情况。结果4种中药单体高浓度亚组间RAW264.7和DC2.4细胞中CD80、CD86、MHCⅠ和MHCⅡ表达水平比较差异均有显著性(F=5.40~44.18,P<0.05);中浓度亚组间DC2.4细胞的4种表面标志物表达水平比较差异均有显著性(F=3.82~170.14,P<0.05)。白杨素组的各亚组间RAW264.7和DC2.4细胞的CD80、CD86、MHCⅠ和MHCⅡ表达水平比较差异均有显著性(F=3.89~51.54,P<0.05);蔓荆子黄素组和刺芒柄花素组的各亚组间RAW264.7细胞的CD80、CD86和MHCⅠ表达水平比较差异有显著性(F=38.02~1543.14,P<0.05),DC2.4细胞的CD80、CD86和MHCⅡ表达水平比较差异有显著性(F=34.52~240.18,P<0.05);北美圣草素组的各亚组间RAW264.7细胞和DC2.4细胞的CD80和CD86表达水平比较差异有显著性(F=76.04~1543.14,P<0.05),DC2.4细胞的MHCⅠ和MHCⅡ表达水平比较差异有显著性(F=9.46、54.12,P<0.05)。结论高浓度的4种中药单体均对RAW264.7和DC2.4细胞系有不同程度的免疫激活作用,其中白杨素对APCs的免疫激活潜能最强。Objective To assess the ability of four traditional Chinese medicine monomers(chrysin,vitexicarpin,for-mononetin,and eriodictyol)to activate antigen-presenting cells(APCs)at the cellular level,and to select monomers with immune activation effects.Methods Based on treatment methods,RAW264.7 cells and DC2.4 cells were divided into chrysin group,vitexicarpin group,formononetin group,and eriodictyol group.Each group was further divided into low-,medium-,and high-concentration subgroups.Negative control subgroups and positive control subgroups(treated with lipopolysaccharide)were set.After 48 h of treatment,flow cytometry was used to measure the expression of CD80,CD86,major histocompatibility complexⅠ(MHCⅠ),and MHCⅡon the surface of RAW264.7 cells and DC2.4 cells in each subgroup.Results CD80,CD86,MHCⅠ,and MHCⅡshowed significantly different expression levels on RAW264.7 and DC2.4 cells between the high-concentration subgroups of the four monomers(F=5.40-44.18,P<0.05),and on DC2.4 cells between the medium-concentration subgroups(F=3.82-170.14,P<0.05).The chrysin subgroups showed significantly different expression levels of CD80,CD86,MHCⅠ,and MHCⅡon RAW264.7 and DC2.4 cells(F=3.89-51.54,P<0.05).For both the vitexicarpin and formononetin subgroups,significant differences were observed in the expression levels of CD80,CD86,and MHCⅠon RAW264.7 cells(F=38.02-1543.14,P<0.05)and CD80,CD86,and MHCⅡon DC2.4 cells(F=34.52-240.18,P<0.05).There were significant differences between the eriodictyol subgroups in the expression levels of CD80 and CD86 on RAW264.7 cells and DC2.4 cells(F=76.04-1543.14,P<0.05)and MHCⅠand MHCⅡon DC2.4 cells(F=9.46,54.12,P<0.05).Conclusion The four traditional Chinese medicine monomers can activate RAW264.7 and DC2.4 cell lines to a varying extent,among which chrysin has the greatest immune activation effects on APCs.
