摘 要:目的:探讨益生菌联合不同强度有氧运动对2型糖尿病大鼠骨骼肌脑源性神经营养因子(BDNF)、核因子-κB(NF-κB)、肾脏脂体比、脂肪量的影响。方法:选取60只清洁级雄性C57/BL大鼠,造模成功后将2型糖尿病大鼠采用随机数字表法分为五组,分别为模型组、A组(益生菌)、B组(益生菌+运动A强度)、C组(益生菌+B强度)、D组(益生菌+C强度)。A组、B组、C组、D组每日灌胃嗜酸乳杆菌1 mL(益生菌数量为1×10^(9) CFU/mL),B组、C组、D组都给予跑台有氧运动训练,分别按10、20、40 m/min速度进行跑台训练,每天训练1 h。五组大鼠治疗观察时间为8周。比较五组大鼠的血糖、胰岛素抵抗水平、肾脏脂肪量和脂体比、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、BDNF、NF-κB水平。结果:治疗4、8周,A组、B组、C组、D组的空腹血糖与胰岛素抵抗水平均低于模型组,且D组均低于A组、B组、C组,C组均低于B组、A组,B组均低于A组,差异均有统计学意义(P<0.05)。治疗8周,A组、B组、C组、D组的肾脏脂肪量和脂体比、血清IL-1β、TNF-α含量、腓肠肌BDNF与NF-κB蛋白相对表达水平均低于模型组,且D组均低于A组、B组、C组,C组均低于B组、A组,B组均低于A组,差异均有统计学意义(P<0.05)。结论:益生菌联合不同强度有氧运动在2型糖尿病大鼠中应用,能抑制腓肠肌BDNF与NF-κB蛋白的表达,还可抑制血清炎症因子的释放,从而改善大鼠的血糖与肾脏脂体比、脂肪量。Objective:To investigate the effects of probiotics combined with different intensities of aerobic exercise on skeletal muscle brain-derived neurotrophic factor (BDNF),nuclear factor-kappa B (NF-κB),and renal lipid-to-body ratio and fat mass in skeletal muscle of type 2 diabetic rats.Method:A total of 60 clean-grade male C57/BL rats were selected,after modeling they were randomly divided into 5 groups,model group,group A (probiotics),group B (probiotics + exercise intensity A),group C (probiotics +exercise intensity B),and group D (probiotics + exercise intensity C).Group A,group B,group C and group D were given 1 mL lactobacillus acidophilus (probiotics amount was 1×10~9 CFU/mL) by intragastric administration.Group B,C and D were given treadmill aerobic exercise training at 10,20 and 40 m/min,respectively,for 1 h every day.The rats in the five groups were treated for 8 weeks.The levels of blood glucose,insulin resistance,renal lipid-to-body ratio and fat mass,interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),BDNF and NF-κB were compared among five groups.Result:After 4 and 8 weeks of treatment,the levels of fast blood glucose and insulin resistance in the group A,group B,group C and group D were lower than those in the model group,and those in the group D were lower than those in the group A,group B and group C,those in the group C were lower than those in the group B and group A,and those in the group B were lower than those in the group A,the differences were statistically significant (P<0.05).After 8 weeks of treatment,the kidney fat content,lipid-to-body ratio,serum IL-1β and TNF-α contents,and the relative expression level of BDNF and NF-κB protein in gastrocnemius muscle in the group A,group B,group C and group D were lower than those in the model group,and those in the group D were lower than those in the group A,group B and group C,those in the group C were lower than those in the groups B and group A,and those in the group B were lower than those in the group A,the differences were statistic
