摘 要:目的:探究不同浓度硼添加对皖西白鹅产蛋期卵巢中IGFBP3、DRD2和CDK1基因表达的影响,为分析硼在皖西白鹅产蛋期的生理作用提供理论依据。方法:采集饲喂含有不同硼添加量日粮的皖西白鹅卵巢组织,每组3只,提取总RNA,检测RNA的完整性,以求获得品质较好的RNA和RNA电泳胶图,进行反转录。筛选IGFBP3、DRD2和CDK1基因为检测对象,通过实时荧光定量PCR检测各组间3个基因的表达水平。结果:与对照组相比,饲料中添加不同浓度的硼可使卵巢中的IGFBP3基因表达极显著上调(P<0.01),DRD2基因表达上调且在添加低浓度硼情况下表达差异显著(P<0.05);添加低浓度硼可使CDK1基因表达极显著上调(P<0.01),添加高浓度硼可使CDK1基因表达显著上调(P<0.05)。结论:添加微量元素硼可以使卵巢中的IGFBP3、DRD2和CDK1基因表达上调,有助于进一步从分子水平探究硼调控皖西白鹅卵巢功能的分子机制。Objective:To explore the effects of different concentrations of boron on the expression of IGFBP3,DRD2 and CDK1 genes in the ovaries of Wanxi white geese during laying period,so as to provide a theoretical basis for analyzing the physiological effect of boron during laying period of Wanxi white geese.Methods:Ovarian tissues were collected from three individuals in each group of Wanxi white geese fedby diets containing different amounts of boron,and total RNA was extracted to detect the integrity of RNA,in order to obtain RNA of good quality and RNA electrophoresis gel maps for reverse transcription.The IGFBP3,DRD2 and CDK1 genes were selected for detection,and the expression levels of these three genes were detected by real-time fluorescence quantitative PCR.Results:Compared with the control group,the addition of different concentrations of boron in the diet caused extreme up-regulation of IGFBP3 gene expression in ovaries(P<0.01),and DRD2 gene expression increased with its expression increasing significantly in the lower concentrations of boron addition group(P<0.05).The addition of low concentrations of boron caused highly significant up-regulation of CDK1 gene expression(P<0.01) and the addition of high concentrations of boron caused its significant upregulation(P<0.05).Conclusion:The addition of trace element boron can upregulate the expression of IGFBP3,DRD2 and CDK1 genes in ovaries,which will help to further explore the molecular mechanism of boron regulating the ovarian performance of Wanxi white geese at the molecular level.
