失眠穴方合脐内环穴针刺对PCPA模型大鼠海马5-HT1AR、5-HT2AR表达的影响

摘　　要：目的:观察针刺失眠穴方加脐内环穴对氯苯丙氨酸(para-chlorophenylalanine,PCPA)诱导下失眠大鼠治疗后海马5-HT_(1A)R、5-HT_(2A)R表达的影响,探讨针刺改善失眠症的机制。方法:取46只健康SPF级雄性SD大鼠,随机抽取10只为正常组,余36只腹腔注射PCPA造模,筛选出30只造模成功大鼠分为模型组、针刺组、非穴组,每组各10只。各组大鼠经相应干预处理6天后,采取灌注分离法取海马组织。采用免疫组化法观察各组5-HT_(1A)R、5-HT_(2A)R的形态学变化,统计5-HT_(1A)R、5-HT_(2A)R平均光密度值。结果:模型组大鼠海马5-HT_(1A)R阳性表达数目下降,5-HT_(2A)R阳性表达数目升高;与模型组比较,针刺组5-HT_(1A)R阳性表达数目增加(P<0.05),5-HT_(2A)R阳性表达数目降低(P<0.05)。与模型组比较,针刺组5-HT_(1A)R平均光密度值升高(P<0.05),5-HT_(2A)R平均光密度值降低(P<0.05)。结论:针刺失眠穴方加脐内环穴针法可良性调节PCPA模型大鼠5-HT_(1A)R、5-HT_(2A)R在海马区的表达,进而起到改善失眠的作用。Objective:To observe the effects of insomnia acupoints prescription and umbilical inner acupoints needling on the expressions of 5-HT_(1A)R and 5-HT_(2A)R in hippocampus of PCPA-induced insomnia rat models,and explore the mechanism of acupuncture improving insomnia.Methods:All 46 healthy SPF male SD rats were chosen,ten were selected as the normal group randomly,the remaining 36 ones were established by intraperitoneal injection of PCPA,30 rat models were screened out and divided into the model group,the acupuncture group and non-acupoint group,ten rats in each group.After intervening for six days in different groups,perfusion separation method was adopted to take hippocampal tissues from rats.Immunohistochemical method was used to observe morphological changes of 5-HT_(1A)R and 5-HT_(2A)R in different groups,and count optical density average of 5-HT_(1A)R and 5-HT_(2A)R.Results:The number of positive expressions of 5-HT_(1A)R of the model group decreased,while the number of positive expressions of 5-HT_(2A)R raised;compared with the model group,the number of positive expressions of 5-HT_(1A)R increased in the acupuncture group(P<0.05),the number of positive expressions of 5-HT_(2A)R reduced(P<0.05).Compared with the model group,optical density average of 5-HT_(1A)R ascended in the acupuncture group(P<0.05),optical density average of 5-HT_(2A)R decreased(P<0.05).Conclusion:Insomnia acupoints prescription and umbilical inner acupoints needling could adjust the expressions of 5-HT_(1A)R and 5-HT_(2A)R in hippocampus of PCPA-induced insomnia rat models better,thereby to improve insomnia.
