摘  要:目的探讨肉苁蓉苯乙醇总苷(CPhGs)对H22荷瘤小鼠肿瘤细胞自噬的影响。方法利用H22肝癌细胞株建立荷瘤小鼠模型,60只雄性昆明小鼠随机分为6组(10只/组):正常对照组(蒸馏水)、模型组(蒸馏水)、模型+阳性对照组(西黄丸780 mg/kg·bw)、模型+CPhGs低剂量组(125 mg/kg·bw)、模型+CPhGs中剂量组(250 mg/kg·bw)、模型+CPhGs高剂量组(500 mg/kg·bw)。除正常对照组以外,其余各组造模后24 h,每组均按设定剂量灌胃给予蒸馏水或受试药物CPhGs、西黄丸,1次/d,连续20 d。利用透射电子显微镜检测自噬小体、免疫组化法定位、Western blot法检测自噬相关蛋白LC3BⅡ和p62。结果透射电子显微镜观察细胞超微结构发现,CPhGs高剂量组自噬小体与自噬溶酶体数量增多;免疫组化及Western blot结果显示,高剂量CPhGs干预后可上调自噬相关蛋白LC3BⅡ表达(P<0.01),下调P62蛋白表达(P<0.01)。结论CPhGs具有诱导肿瘤细胞自噬的作用。Objective To explore the effect of CPhGs on autophagy of tumor cells in H22 tumor-bearing mice.Methods H22 hepatoma mouse models were established.Sixty male Kunming mice were randomly divided into six groups(10 mice/group):normal control group(distilled water),model group(distilled water),model+positive control group(Xihuang Wan 780 mg/kg·bw),model+CPhGs low dose group(125 mg/kg·bw),model+CPhGs medium dose group(250 mg/kg·bw),model+CPhGs high dose group(500 mg/kg·bw).Except for the normal control group,the rest of the groups were given distilled water or the test drug CPhGs and Xihuang Wan by gavage at the set dose on the second day after modeling,once a day for 20 consecutive days.Transmission electron microscopy was used to detect autophagosomes,immunohistochemical localization.Western blot method was used to detect autophagy-related proteins LC3 BⅡand p62.Results Transmission electron microscopy observation of cell ultrastructure revealed that the number of autophagosomes and autophagolysosomes increased in the high-dose CPhGs group;The result of immunohistochemistry and Western blot showed that high-dose CPhGs can up-regulate the expression of autophagy-related protein LC3 BⅡ(P<0.01)and down-regulate the expression of P62 protein(P<0.01).Conclusion CPhGs can induce autophagy in tumor cells.
