活络骨康丸对兔酒精性股骨头坏死骨特异性碱性磷酸酶和骨钙素的影响

摘　　要：目的:探讨活络骨康丸治疗兔股骨头坏死对成骨指标骨特异性碱性磷酸酶(BALP)和骨钙素(BGP)的影响,分析药物作用靶细胞。方法:将30只日本大耳白兔随机分为实验组、模型对照组和空白对照组,每组10只。实验组和模型对照组给予白酒(酒精度为46%vol)10 mL·kg^(-1)·d^(-1)灌胃90 d后,实验组停止白酒灌胃,改用活络骨康丸0.84 g·kg^(-1)·d^(-1)灌胃60 d;模型对照组改为正常饲养60 d。空白对照组给予质量分数为10%的葡萄糖溶液10 mL·kg^(-1)·d^(-1),持续灌胃90 d后,改为正常饲养60 d。150 d后,3组静脉采血,测定BALP和BGP浓度,取双侧股骨头显微镜下观察骨组织。结果:显微镜下可见实验组骨小梁连续、模型对照组骨小梁大量破坏。实验组BALP和BGP浓度值均高于模型对照组,差异有统计学意义(P<0.05)。结论:活络骨康丸治疗酒精性股骨头坏死,能够抑制骨髓间充质干细胞成脂化,促进成骨细胞功能,分泌合成BALP和BGP。Objective:To investigate the changes of bone-specific alkaline phosphatase(BALP)and bone gla protein(BGP)in rabbits with osteonecrosis of femoral head treated by Huoluo Gukang Wan(活络骨康丸),and to analyze the target cells of drugs.Methods:Thirty Japanese white rabbits were randomly divided into an experimental group,a model control group and a blank control group,with 10 rabbits in each group.The experimental group and the model group were given 10 mL·kg^(-1)·d^(-1) of strong liquor(containing 46%alcohol,vol).After 90 days,liquor for the experimental group was changed into Huoluo Gukang Wan(0.84 g·kg^(-1)·d^(-1))for 60 days,the model group was normally fed for 60 days.The blank control group was given 10 mL·kg^(-1)·d^(-1) of 10%glucose solution by gavage for 90 days,and then changed to normal feeding for 60 days.After 150 days,blood samples were taken from 3 groups and the concentrations of BALP and BGP were measured.The bilateral femoral heads were taken and the bone tissues were observed under the microscope.Results:Under the microscope,the trabeculae in the experimental group were continuous,and the trabeculae in the model group were destroyed.The concentrations of BALP and BGP in the experimental group were higher than those in the model group(P<0.05).Conclusion:Huoluo Gukang Wan can treat alcoholic necrosis of femoral head,inhibit the adipogenesis of bone marrow mesenchymal stem cells,promote the function of osteoblasts,and secrete BALP and BGP.
