摘 要:【目的】确定药用植物路边青(Geum aleppicum Jacq.)叶绿体基因组结构、特征及基因组成情况,比较其与近缘种的叶绿体基因组结构和系统发育的关系。【方法】以路边青为材料,利用高通量技术测定基因组DNA序列,用NOVO Plasty组装叶绿体基因组,并进行序列分析,采用最大似然法(maximum likelihood ML)构建系统进化树。【结果】(1)路边青叶绿体基因组为典型的4分区域结构,全长为155911 bp,GC值为35.6%,包括1个大单拷贝区(LSC)、1对反向重复区(IR)和1个短单拷贝区(SSC),序列长度分别为85384、26119、18298 bp。路边青的叶绿体基因组共有132个基因,其中编码蛋白基因、rRNA基因与tRNA基因的数量分别为87、8、37个;(2)根据NCBI公开发布的24个蔷薇亚科植物的叶绿体全基因组信息,筛选出蔷薇亚科植物叶绿体基因组的14个变异位点较高的叶绿体基因间隔区;(3)系统进化分析表明,路边青与同属的Geum macrophyllum Willd.和Geum rupestre(T.T.Yu&C.L.Li)Smedmark.三者构成一单系分支,具有100%的支持率。【结论】探究了药用植物路边青的叶绿体全基因组,并基于叶绿体基因组数据再次验证,药用植物路边青隶属于蔷薇亚科的路边青属(Geum L.),与该属的Geum macrophyllum Willd.和Geum rupestre(T.T.Yu&C.L.Li)Smedmark.亲缘关系较近。【Objective】The study was conducted to determine the chloroplast genome structure,characteristics and gene composition of Geum aleppicum Jacq.,and to compare its relationship with chloroplast genome structure and phylogeny of related specie.【Methods】Geum aleppicum Jacq.was used as material to determine its genomic DNA sequence using high-throughput technique.The chloroplast genome was assembled using NOVO,and sequence analysis was performed.The phylogenetic tree was constructed using Maximum-Likelihood(ML)methods.【Results】(1)The results showed that the chloroplast genome of Geum aleppicum Jacq.was 155911 bp in length with a GC content of 35.6%.The chloroplast genome exhibited a typical quadripartite structure,including a large single copy region(LSC),a pair of inverted repeats(IR),and a small single copy(SSC),and the sequence lengths were 85384 bp,26119 bp,and 18298 bp.The chloroplast genome harbored 132 genes,including 87 protein-coding genes,8 rRNA genes,and 37 tRNA genes.(2)Based on the chloroplast genome information of 24 Rosoideae plants released publicly by NCBI,we screened out 14 spacer genes with higher mutation sites.(3)Phylogenetic analysis showed that Geum aleppicum Jacq.,Geum macrophyllum Willd.and Geum rupestre(T.T.Yu&C.L.Li)Smedmark of the genus formed a single branch with 100 percent supporting rate.【Conclusion】In this study,we first explore the complete chloroplast genome of traditional Chinese medicinal Geum aleppicum Jacq..Our result verified that Geum aleppicum Jacq.belonged to the Rosoideae,and was related to Geum macrophyllum Willd.and Geum rupestre(T.T.Yu&C.L.Li)Smedmark.
