摘 要:利用荧光标记毛细电泳技术对34个重瓣萱草品种进行了SSR分子标记检测,分析其遗传多样性,并筛选出12对多态引物对供试材料重瓣萱草进行了PCR扩增。结果表明,12对引物共获得111个等位基因,平均9.2500个,其SSR位点的遗传多样性变化范围为0.5826~0.9277,杂合度变化范围为0.1471~0.7647,多态性信息含量变化范围为0.5684~0.9236。UPGMA聚类分析图显示,当遗传相似系数为0.4182时,可将34个重瓣萱草品种分为两大类群,第一类以‘中国山’‘双面娇娃’‘粉红女孩’‘双重美味’为代表,共4个重瓣萱草品种;第二类以‘残酷的命运’‘粉娃娃’‘摩西火’‘双子座侍者’‘四十二街’等为代表,共30个重瓣萱草品种。说明这34个重瓣萱草品种具有较高的遗传多样性,遗传变异类型丰富。本研究结果可为今后培育更多的重瓣萱草新品种提供参考和依据。The fluorescent labeling capillary electrophoresis technology was used to detect their genetic diversity of 34 double-petaled Hemerocallis varieties by SSR molecular markers.Twelve pairs of polymorphic primers were screened for PCR amplification of the test material Hemerocallis.The results showed that a total of 111 alleles were obtained from the 12 pairs of primers,with an average of 9.2500,and the genetic diversity of the SSR loci ranged from 0.5826 to 0.9277.The range of heterozygosity was 0.1471~0.7647,and the range of polymorphism information content was 0.5684~0.9236.The UPGMA cluster analysis chart shows that according to the genetic similarity coefficient of 0.4182,34 double-petaled Hemerocallis varieties can be divided into two major groups.‘Yummy’represents a total of 4 double-petaled Hemerocallis varieties.The second category is 30 double-petaled Hemerocallis varieties such as‘Cruel Destiny’,‘Pink Doll’,‘Moses Fire’,‘Gemini Waiter’and‘Forty-second Street’.This shows that SSR molecular markers can completely distinguish these 34 double-petal daylilies,which will provide reference and basis for the cultivation of more double-petaled daylilies in the future.
