复方丹参注射液对家兔退变腰椎间盘的TNF-α与CTGF的表达影响

摘　　要：目的:研究家兔退变性椎间盘内注射复方丹参注射液对椎间盘组织中的TNF-α与CTGF影响。方法:将36只健康成年家兔随机分为观察组、模型组、空白组,每组12只。观察组和模型组分别用针刺法建立椎间盘退变动物模型,观察组注射复方丹参注射液,空白组不进行手术干预,并分别于第2周、第4周、第6周、第12周将家兔处死获得椎间盘标本。分别进行HE染色和免疫组化技术处理。对观察组、模型组与空白组的椎间盘组织中的TNF-α和CTGF的阳性表达分别进行定性、定位与半定量检测,并对TNF-α和CTGF的阳性表达率进行分析。结果:模型组与空白组比较,TNF-α与CTCF表达明显增加;观察组与模型组比较TNF-α与CTCF表达明显减少。结论:椎间盘内注射复方丹参注射液,对于家兔退变椎间盘中的TNF-α与CTGF表达有抑制作用,能够抑制椎间盘退行性病变的进展。Objective:To study the effect of Danshen injection on TNF in degenerative intervertebral disc of rabbits-αAnd CTGF.Methods:36 healthy adult rabbits were randomly divided into experimental group,model group and blank group,with 12 rabbits in each group.The animal models of intervertebral disc degeneration were established by Acupuncture in the experimental group and the model group respectively.The experimental group was injected with Salvia miltiorrhiza injection,and the blank group did not undergo surgical intervention.The rabbits were killed at the 2nd,4th,6th and 12th weeks respectively to obtain intervertebral disc samples.He staining and immu-nohistochemistry were performed.TNF in intervertebral disc tissue of experimental group,model group and blank group-αAnd CTGF were detected qualitatively,regionally and semi quantitatively,and TNF was detected-αThe positive expression rates of CCN2 and CCN2 were analyzed.Results:Compared with the blank group,TNF-α The expression of and CTGF increased significantly;TNF was compared between experimental group and model group-αAnd the expression of CTGF decreased significantly.Conclusion:intradiscal injection of Salvia miltiorrhiza injection has no effect on TNF in degenerative intervertebral discs of rabbits-α It can inhibit the expression of CTGF and inhibit the progression of intervertebral disc degeneration.
