摘 要:目的探究程序性坏死与氟诱导的人成骨细胞氧化损伤的关系。方法选择人成骨肉瘤细胞系(Saos-2)体外培养,并用不同浓度的氟化钠(0、5、10、20、40 mg/L)干预以建立氟致成骨细胞损伤模型;利用分光光度法测定细胞中T-SOD、GSH、MDA的含量。结果实验组中T-SOD的活力及GSH的含量均低于相同染氟条件下的对照组(P<0.05);T-SOD的活力及GSH的含量随处理浓度的增加而下降,两者存在明显负相关(-1<r<0,P<0.05);干预时间对于细胞中T-SOD的活性和GSH的含量的影响存在双向性,即在氟干预浓度小于20 mg/L时,T-SOD的活力和GSH的含量与干预时间呈正相关,在氟干预浓度大于20 mg/L时,T-SOD的活力及GSH的含量与干预时间呈负相关;实验组中MDA的含量均高于相同染氟条件下的对照组(P<0.05);MDA的含量随着氟诱导浓度的增加而增加,两者存在明显正相关(0<r<1,P<0.05);在实验组中,干预时间对于细胞中MDA的含量的影响存在单向性,细胞中MDA的含量与干预时间呈正相关,而在对照组中,出现了与T-SOD和GSH相反的趋势。结论氟诱导过程中成骨细胞程序性坏死发生与氧化损伤有关。Objective To explore whether necroptosis exists in the process of fluoride-induced human osteoblast injury.Methods The Saos-2 was cultured in vitro and intervened with different concentrations of sodium fluoride(0,5,10,20,40mg/L)to establish the model of osteoblast injury induced by fluoride.The contents of T-SOD,GSH and MDA in cells were determined by spectrophotometry.Results The activity of T-SOD and the content of GSH in the experimental group were lower than those in the control group under the same fluoride exposure(P<0.05),and the activity of T-SOD and the content of GSH decreased with the increase of treatment concentration,and there was a significant negative correlation between them(-1<r<0,P<0.05).The effects of intervention time on the activity of T-SOD and the content of GSH in cells were bi-directional,that is,when the concentration of fluoride intervention was less than 20mg/L,the activity of T-SOD and the content of GSH were positively correlated with the intervention time,while when the concentration of fluoride intervention was higher than that of 20mg/L,the activity of T-SOD and the content of GSH were negatively correlated with the intervention time,The content of MDA in the experimental group was higher than that in the control group under the same fluoride exposure condition(P<0.05);the content of MDA increased with the increase of the fluorine-induced concentration,and there was a significant positive correlation between the two(0<r<1,P<0.05);In the experimental group,the effect of intervention time on the content of MDA in cells was unidirectional,and the content of MDA in cells was positively correlated with the intervention time,while in the control group,there was an opposite trend to T-SOD and GSH.Conclusion The occurrence of programmed necrosis of osteoblasts during fluoride induction is related to oxidative damage.
