摘 要:目的:探究活血消瘿方调控Fas系统对结节性甲状腺肿大鼠模型甲状腺细胞凋亡的影响。方法:将60只SD大鼠随机分为空白对照组、模型组、阳性对照组、活血消瘿方低剂量组、活血消瘿方中剂量组、活血消瘿方高剂量组,每组10只。空白对照组大鼠灌胃生理盐水,其余各组大鼠灌胃0.1%PTU溶液复制结节性甲状腺肿模型。给予相应药物干预4周后,ELISA法检测血清中促甲状腺激素(TSH)、游离三碘甲腺原氨酶(FT_(3))、游离甲状腺素(FT_(4))水平;测定甲状腺组织质量,HE染色分析甲状腺组织的病理学变化;TUNEL分析甲状腺细胞的凋亡指数;免疫组织化学、Western blotting和RT-PCR检测FasL、Fas、Caspase-8、Caspase-3蛋白和mRNA表达水平。结果:模型组大鼠血清中TSH含量明显高于空白对照组(P<0.05),FT_(3)和FT_(4)含量、甲状腺组织质量、凋亡指数及FasL、Fas、Caspase-8、Caspase-3蛋白和mRNA表达水平均低于空白对照组(P<0.05);阳性对照组和活血消瘿方低、中、高剂量组大鼠血清中TSH含量明显低于模型组(P<0.05),FT_(3)和FT_(4)含量、甲状腺组织质量、凋亡指数及FasL、Fas、Caspase-8、Caspase-3蛋白和mRNA表达水平均明显高于模型组(P<0.05);活血消瘿方低、中剂量组大鼠血清中TSH含量高于阳性对照组(P<0.05),FT_(3)和FT_(4)含量、甲状腺组织质量、凋亡指数及FasL、Fas、Caspase-8、Caspase-3蛋白和mRNA表达水平均明显低于阳性对照组(P<0.05);活血消瘿方高剂量组大鼠以上各指标与阳性对照组比较,差异均无统计学意义(P>0.05)。结论:活血消瘿方能通过激活Fas系统促进结节性甲状腺肿大鼠模型甲状腺细胞凋亡。Objective:To explore the effect of Huoxue Xiaoying Recipe on thyroid cell apoptosis in rat models of nodular goiter by regulating Fas system.Methods:60 SD rats were randomly divided into blank control group,model group,positive control group,and low,medium,and high dose Huoxue Xiaoying Recipe groups,10 in each group.Rats in the blank control group were gavaged with normal saline,and rats in other groups were gavaged with 0.1%PTU solution to replicate the nodular goiter model.After 4 weeks of intervention with corresponding drugs,the levels of thyroid stimulating hormone(TSH),free triiodothyronine aminotransferase(FT_(3))and free thyroxine(FT_(4))in serum were detected by ELISA.The quality of thyroid tissue was measured,and the pathological changes of thyroid tissue were analyzed by HE staining.TUNEL was used to analyze the apoptosis index of thyroid cells.The expression levels of FasL,Fas,Caspase-8,Caspase-3 protein and mRNA were detected by immunohistochemistry,Western blotting and RT-PCR.Results:The serum levels of TSH in the model group were higher than those in the blank control group(P<0.05),while the contents of FT_(3) and FT_(4),thyroid tissue weight,apoptosis index,and the expression levels of FasL,Fas,Caspase-8,Caspase-3 and mRNA were lower than those in the blank control group(P<0.05).The content of TSH in serum of rats in the positive control group and the low,medium,and high dose groups of Huoxue Xiaoying Recipe were significantly lower than that in model group(P<0.05),and the contents of FT_(3) and FT_(4),the weight of thyroid tissue,apoptosis index and the levels of FasL,Fas,Caspase-8,Caspase-3 and mRNA in the model group were significantly higher than those in the model group(P<0.05).The content of TSH in serum of rats in the low and medium dose groups was higher than those in the positive control group(P<0.05),and the contents of FT_(3) and FT_(4),the weight of thyroid tissue,apoptosis index,and the expression levels of FasL,Fas,Caspase-8,Caspase-3 and mRNA were significantly lower than those in t
