摘 要:目的:探究电针对脑梗死大鼠脑组织中脑源性神经营养因子/酪氨酸激酶/磷脂酰肌醇3激酶(BDNF/TrkB/PI3K)通路的影响。方法:采用改良线栓法构建脑梗死大鼠模型,将建模成功的大鼠随机分为模型组、电针组、抑制剂组、电针+抑制剂组,每组15只,另随机取15只大鼠作为假手术组;抑制剂组、电针+抑制剂组大鼠经右侧脑室注射10μg/kg K252a,电针组、电针+抑制剂组大鼠对百会、风府、心俞、内关4个穴位进行电针干预,1次/d,连续干预7 d。干预结束后,对各组大鼠进行神经功能缺损评分;2,3,5-三苯基氯化四氮唑(TTC)染色法分析大鼠脑梗死面积;酶联免疫吸附(ELISA)法检测大鼠血清IL-8、TNF-ɑ、IL-2水平;Western blotting法检测大鼠脑组织中BDNF/TrkB/PI3K通路相关蛋白表达。结果:与假手术组比较,模型组大鼠神经功能缺损评分、脑梗死面积及血清IL-8、IL-2、TNF-ɑ含量均明显升高(P<0.05),BDNF、TrkB蛋白相对表达量及p-PI3K/PI3K、p-AKT/AKT均明显降低(P<0.05)。与模型组比较,电针组大鼠神经功能缺损评分、脑梗死面积及血清IL-8、IL-2、TNF-ɑ含量均明显降低(P<0.05),BDNF、TrkB蛋白相对表达量及p-PI3K/PI3K、p-AKT/AKT均明显升高(P<0.05);抑制剂组大鼠神经功能缺损评分、脑梗死面积及血清IL-8、IL-2、TNF-ɑ含量均明显升高(P<0.05),BDNF、TrkB蛋白相对表达量及p-PI3K/PI3K、p-AKT/AKT均明显降低(P<0.05);电针+抑制剂组大鼠各指标与模型组比较,差异均无统计学意义(P>0.05)。与电针组比较,电针+抑制剂组大鼠神经功能缺损评分、脑梗死面积及血清IL-8、IL-2、TNF-ɑ含量均明显升高(P<0.05),BDNF、TrkB蛋白相对表达量及p-PI3K/PI3K、p-AKT/AKT均明显降低(P<0.05)。结论:电针可通过激活BDNF/TrkB/PI3K信号通路,减轻脑梗死大鼠脑组织炎症反应,改善神经功能。Objective:To observe the effects of electroacupuncture on the brain-derived neurotrophic factor/tyrosine kinase/phosphatidylinositol 3-kinase(BDNF/TrkB/PI3K)signaling pathway in the brain tissue of rats with cerebral infarction.Methods:A rat model of cerebral infarction was established by the modified suture method,and the successfully modeled rats were randomly divided into model group,electroacupuncture group,inhibitor group,electroacupuncture+inhibitor group,with 15 rats in each group.Another 15 rats were randomly selected as the sham operation group.Rats in the inhibitor group and electroacupuncture+inhibitor group were injected with 10μg/kg K252a through the right ventricle,and the rats in the electroacupuncture group and electroacupuncture+inhibitor group were treated with 4 acupoints of Baihui(CV20),Fengfu(CV16),Xinshu(BL15)and Neiguan(PC6).Electroacupuncture intervention was performed once a day for 7 consecutive days.After the intervention,the rats in each group were scored for neurological deficit.The area of cerebral infarction was analyzed by 2,3,5-triphenyltetrazolium chloride(TTC)staining.Serum IL-8,TNF-ɑ and IL-2 levels were detected by ELISA.The expression of BDNF/TrkB/PI3K pathway related proteins in rat brain was detected by Western blotting.Results:Compared with the sham-operated group,the neurological deficit score,cerebral infarction area,and serum IL-8,IL-2,and TNF-ɑ levels in the model group were significantly increased(P<0.05),and the relative expressions of BDNF and TrkB proteins and p-PI3K/PI3K and p-AKT/AKT were significantly decreased(P<0.05).Compared with the model group,the neurological deficit score,cerebral infarction area,and serum IL-8,IL-2,and TNF-ɑ levels in the electroacupuncture group were significantly decreased(P<0.05),and the relative expressions of BDNF and TrkB proteins and p-PI3K/PI3K and p-AKT/AKT were significantly increased(P<0.05).Compared with the model group,the neurological deficit score,cerebral infarction area,and serum IL-8,IL-2,and TNF-ɑ levels in the in
