摘  要:目的筛选瓜子金抗老年痴呆的有效部位。方法瓜子金原药材经提取分离得不同极性部位,以此为实验材料,通过测定其体外清除1,1-二苯基-2-三硝基苯肼(DPPH)、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二胺盐(ATBS)自由基能力、还原能力及总酚含量评价其抗氧化活性;建立东莨菪碱(3 mg·kg^(-1))致小鼠学习记忆障碍模型及5%D-半乳糖致小鼠亚急性衰老模型,采用水迷宫法观察其对模型小鼠学习记忆能力的影响。结果抗氧化实验:瓜子金乙酸乙酯部位、正丁醇部位体外均能清除自由基,二者均具有抗氧化活性。改善东莨菪碱致小鼠学习记忆障碍实验:阳性药组和瓜子金乙酸乙酯中、高剂量组及正丁醇中、高剂量组小鼠潜伏时间明显缩短(P<0.05,P<0.01);阳性药组、乙酸乙酯中剂量组及正丁醇低、中剂量组能明显增加小鼠跨越平台次数(P<0.05,P<0.01)。抗半乳糖所致小鼠衰老实验:瓜子金正丁醇部位高剂量组能显著降低亚急性衰老小鼠潜伏时间、增加跨越平台次数(P<0.01),能改善亚急性衰老模型小鼠学习记忆能力。结论瓜子金乙酸乙酯、正丁醇部位具有一定的抗老年痴呆生物活性,该结果对寻找瓜子金抗老年痴呆的物质基础,扩大其临床应用范围,发掘瓜子金科学内涵,具有重要意义。Objective To screen effective parts of Polygala japonica Houtt for anti-dementia.Methods The separation was carried out in accordance with the different polar parts of Polygala japonica Houtt.Antioxidant activities of different polar parts of Polygala japonica Houtt in vitro were evaluated by determining DPPH radical scavenging ability,ABTS radical scavenging ability,reduction of ferric ions and total phenolic content.Learning and memory impairment induced by scopolamine(3 mg·kg^(-1))in mice and mice subacute aging model induced by 5%D-galactose were established.The water maze method was adopted to observe the effect of different polar parts of Polygala japonica Houtt on learning and memory ability of model mice.Results Antioxidant experiment:Ethyl acetate extract and n-butanol extract of Polygala japonica Houtt can remove free radicals in vitro.All of them showed antioxidant activity.Effects of different polar parts of Polygala japonica Houtt on improving scopolamine-induced learning and memory impairment in mice:The latency of mice in the medium-,high-dose group of ethyl acetate extract and the medium-dose group of n-butanol extract was significantly shortened(P<0.01).The latency of the mice in the positive group and the n-butanol high-dose group was significantly shortened(P<0.05).The number of mice crossing the platform in the positive group,the medium-dose group of ethyl acetate extract and the low-dose group of n-butanol extract was significantly increased(P<0.01).The number of mice crossing the platform in the medium-dose group of n-butanol extract was also significantly increased(P<0.05).Effects of different polar parts of Polygala japonica Houtt on D-galactose-induced aging mice:The high-dose group of n-butanol extract can significantly reduce the latency of subacute aging mice,increase the number of crossing platforms(P<0.01),and improve the learning and memory ability of subacute aging model mice.Conclusion Ethyl acetate extract and nbutanol extract of Polygala japonica Houtt may have a certain anti-A
