摘  要:目的探究BMP-2/bFGF双基因转染BMSCs复合生物陶瓷骨对兔桡骨缺损的修复作用及可能机制。方法分离培养兔BMSCs并鉴定,将BMP-2/bFGF过表达慢病毒转染兔BMSCs,Western blot验证转染效率;ALP试剂盒检测桡骨组织ALP活性,HE染色观察桡骨组织形态变化,并通过Lane-Sandhu组织学评分分析桡骨修复情况;免疫荧光检测桡骨组织Col I、OC和OPN表达;Western blot方法检测桡骨骨膜VEGF和EGF水平。结果BMP-2/bFGF双基因转染BMSCs改善缺损桡骨组织形态,增加桡骨组织Lane-Sandhu组织学评分,增加桡骨组织Col I、OC和OPN表达及桡骨骨膜VEGF和EGF表达水平。结论BMP-2/bFGF双基因转染BMSCs复合生物陶瓷骨对兔桡骨缺损存在修复作用,其机制可能与增加骨组织VEGF和EGF水平有关。Objective To investigate the repair effect and possible mechanism of BMSCs composite bioceramic bone transfected with BMP-2/bFGF double gene on radial defect in rabbits.Methods Rabbit BMSCs were isolated and cultured,and then was identified by flow cytometry.BMP-2/bFGF overexpressing lentivirus was transfected into rabbit BMSCs and the transfection efficiency was verified by Western blot.ALP activity was detected by ALP kit.Morphological changes of radial tissue were observed by HE staining,and the repair of the radius was analyzed by Lane-Sandhu histological score.The expression of COL I,OC and OPN in radial tissue was detected by immunofluorescence.The levels of VEGF and EGF in radial periosteum were detected by Western blot.Results BMP-2/bFGF double gene transfection of BMSCs improved the morphology of radial tissue defects,increased the Lane-Sandhu histological score of radial tissue,increased the expression of COL I,OC and OPN in radial tissue,and increased the levels of VEGF and EGF in radial periosteum.Conclusion BMP-2/bFGF double gene transfection of BMSCs composite bioceramics can repair radial defect in rabbits,and the mechanism may be related to the increase of VEGF and EGF levels in bone tissue.
