摘 要:目的探讨人参皂苷CK对色素沉着绒毛结节性滑膜炎成纤维样滑膜细胞(PVNS-FLS)体外活性的影响。方法将细胞分为阴性对照组(Control组)和实验组(CK药物组),MTT与CCK-8方法检测不同浓度药物处理对细胞增殖能力的影响;倒置显微镜下观察各组细胞形态学变化;Hoechst 33342及TUNEL凋亡染色试剂盒检测不同浓度药物处理对PVNS-FLS的诱导凋亡作用。结果不同浓度药物作用细胞后,PVNS-FLS的体外生存率及体外增殖能力显著下降,并呈明显的浓度依赖性及时间依赖性(P<0.05);Hoechst 33342及TUNEL凋亡染色检测结果证实,CK能诱导PVNS-FLS凋亡(P<0.05)。结论人参皂苷CK能抑制PVNS-FLS的体外活性及增殖能力,并诱导凋亡。Objective To explore the effect of ginsenoside CK on the in vitro activity of fibroblast-like synovial cells(PVNS-FLS)in pigmented villonodular synovitis.Methods The cells were divided into negative control group(Control group)and experimental group(CK drug group).MTT and CCK-8 methods were used to detect the effects of different concentrations of drugs on cell proliferation.The morphological changes of cells in each group were observed under inverted microscope.Hoechst 33342 and TUNEL apoptosis staining kit were used to detect the apoptosis-inducing effects of different concentrations of drugs on PVNS-FLS.Results In vitro survival rate and proliferation ability of PVNS-FLS were significantly decreased after treatment with different concentrations of pvNS-FLS,and showed significant concentration and time dependence(P<0.05).Hoechst 33342 and TUNEL staining confirmed that CK could induce pvNS-FLS apoptosis(P<0.05).Conclusion Ginsenoside CK could inhibit pvNS-FLS proliferation and induce apoptosis in vitro.
