摘 要:目的:通过对革兰染色液配方的改良,建立一种快速两步革兰染色法.方法:配制多种两步法革兰染色液,不断优化染色时间,建立一种快速两步革兰染色法,然后将其与临床快速四步革兰染色法染色结果进行比较.结果:两步法A液15s、B液10s染色效果好,分别用两步法、四步法各染100张涂片,其中两步法合格率为94%,四步法为87%.以合格率计算,二者结果经统计学分析无明显差异,两步法可以达到与现有临床快速四步法相同的染色效果.结论:本研究建立的两步革兰氏法能满足临床工作需要,可以用于临床.Objective To establish a fast two-step Gram staining method by improving the formula of Gram staining solution.Methods A variety of two-step Gram staining solutions were prepared,which can continuously optimize the staining time,and a fast two-step Gram staining method was established,and then the staining results were compared with the clinical fast four-step Gram staining method.Results Two-step staining with solution A for 15s and solution B for 10s was effective,and 100 smears were dyed with two-step method and four-step method respectively,among which the qualified rate of two-step method was 94%and that of four-step method was 87%.Calculated by the qualified rate,there was no significant difference between the two results by statistical analysis.The two-step method can achieve the same staining effect as the existing clinical fast four-step method.Conclusion Two-step Gram method established in this study can meet the needs of clinical work and can be used in clinic.
