摘 要:目的:建立了产术康合剂HPLC-DAD法指纹图谱分析方法,采用血清药物化学的研究方法,对产术康合剂大鼠口服后的入血成分进行分析.方法:采用ACE EXCEL C18-AR色谱柱(250 mm×4.6 mm,5μm),以0.1%磷酸水溶液-乙腈为梯度洗脱的流动相,体积流量为1.0 mL/min,柱温为35℃,检测波长为215 nm.采用"中药色谱指纹图谱相似度评价系统软件(版本2004A)"分别对10批产术康合剂的指纹图谱进行相似度评价,并将对照图谱与各单味药材进行色谱峰匹配,确定共有峰归属,结合混合标准溶液图谱对共有峰进行成分指认.通过SD大鼠灌胃给予产术康合剂,采用HPLC-DAD分析其含药血样色谱图,通过与空白血清的色谱图对比分析,探讨产术康合剂的体内移行成分.结果:10批产术康合剂指纹图谱与对照图谱的相似度大于0.96,共有26个峰,所有共有峰都能归属到单味药材.含产术康血清与空白血清比较,发现8个色谱峰,其中7种为原型成分入血,1种可能是产术康合剂在大鼠体内产生的代谢物.结论:建立了产术康合剂的HPLC指纹图谱,初步明确了其活性物质基础,为该制剂的质量标准优化提供了科学依据,且为产术康合剂的药动学研究奠定了基础.Objective The HPLC-DAD fingerprint analysis method of Chanshukang Mixture was established,and the blood components of Chanshukang Mixture after oral administration were analyzed using the research method of serum medicinal chemistry.Methods The separation was performed on An ACE EXCEL Cjs-AR(250 mm x 4.6 mm,5/im)with gradient elution of acetonitrile—0.1%phosphoric acid.And the flowing rate was 1.0 mI7min.The column temperature was 35 Tl and the detection wavelength was 215 nm.The Chinese Medicine Chromatography Fingerprint Similarity Evaluation System Software(Version 2004A)was used to evaluate the similarity degree of the fingerprints of 10 batches of Chanshukang mixture,respectively.The chromatographic peaks of the control spectrum and the single medicinal material were matched to determine the attribution of common peak,and the components of common peak were identified by mixing standard solution.SD rats were intragastrically administered Chanshukang mixture,and the chromatogram of the drug-containing blood sample was analyzed by HPLC-DAD.The chromatogram of the blood sample was compared with that of blank serum to explore the internal migration components of Chanshukang mixture.Results rrhe similarity between the fingerprint patterns of 10 batches of Chanshukang mixture and the control spectra was over than 0.96.There were 26 peaks in total,and all the common peaks could be attributed to a single medicinal material.Comparing the serum containing Chanshukang with the blank serum,8 chromatographic peaks were found,of which 7 were prototype components into the blood,and one might be a metabolite produced by Chanshukang mixture in rats.Conclusion The HPLC fingerprint of Chanshukang mixture has been established,and its active substance basis has been clarified preliminarily,which provides a scientific basis for the optimization of the quality standard of the preparation,and lays a foundation for the pharmacokinetic study of Chanshukang mixture.
