摘  要:目的探讨葛根对结扎冠状动脉左前降支致心肌缺血大鼠的作用及初步机制。方法将72只SD大鼠随机分为假手术组、模型组、维拉帕米(4 mg·kg^(-1))组及葛根低、中、高剂量组(0.79、1.58、3.15 g·kg^(-1)),每组12只。灌胃给药,每日1次,连续30 d。采用结扎大鼠冠状动脉左前降支复制心肌缺血模型。检测心肌缺血大鼠Ⅱ导联心电图参数;采用伊文思蓝染色法测定大鼠心肌组织缺血面积;检测大鼠血清乳酸脱氢酶(LDH)、天门冬氨酸氨基转移酶(AST)、肌酸激酶(CK)的含量;采用HE染色法观察大鼠心肌组织病理变化;采用Western Blot法检测心肌组织中Bax、Bcl-2蛋白表达。结果与假手术组比较,模型组大鼠心电图ST段抬高幅度显著上升(P<0.01),心肌缺血区面积显著增大(P<0.01),血清AST、LDH、CK含量明显升高(P<0.05,P<0.01),组织病理学检测显示心肌损伤明显加重,心肌组织Bax蛋白表达水平显著升高(P<0.01),Bcl-2蛋白表达水平及Bcl-2/Bax比值显著降低(P<0.01)。与模型组比较,葛根组大鼠心电图ST段抬高幅度均明显降低(P<0.01),心肌缺血区面积显著减少(P<0.01),血清AST、LDH、CK含量均明显下降(P<0.05,P<0.01),组织病理学检测显示心肌损伤明显减轻,心肌组织Bax蛋白表达水平显著降低(P<0.01),Bcl-2蛋白表达水平及Bcl-2/Bax比值显著升高(P<0.01)。结论葛根对结扎冠状动脉左前降支致心肌缺血大鼠有一定的保护作用,其作用机制可能与调节Bcl-2/Bax凋亡信号分子有关。Objective To investigate the effect and preliminary mechanism of Radix Puerariae on myocardial ischemia induced by ligated left anterior descending(LAD)coronary artery.Methods Seventy-two SD rats were randomly divided into sham-operated group,model,Verapamil(4 mg·kg^(-1))and Radix Puerariae low-,mediumand high-dose groups(0.79,1.58 and 3.15 g·kg^(-1)),12 rats in each group.The myocardial ischemia model was replicated by ligating the LAD coronary artery in rats;The parameters of lead ECG of rats with myocardial ischemia were examined;The rats were stained with Evans Blue(EB)staining to determine the ischemic area of myocardial tissues;the levels of serum lactate dehydrogenase(LDH),aspartate aminotransferase(AST)and creatine kinase(CK)were measured;the histopathological changes of myocardial tissues were observed by HE staining;the protein expressions of Bax and Bcl-2 in myocardial tissues was measured by the Western Blot method.Results Compared with the sham-operated group,the elevation of ST segment in model group was significantly increased(P<0.01),ischemia area was significantly increased(P<0.01),serum contents of AST,LDH and CK were significantly increased(P<0.05,P<0.01),histopathological examination showed that myocardial injury was significantly aggravated.The protein expression of Bax was significantly increased(P<0.01),the protein expressions of Bcl-2,and Bcl-2/Bax ratio were significantly decreased(P<0.01).Compared with the model group,the elevation of ST segment of ECG in the Radix Puerariae group was significantly decreased(P<0.01),the area of myocardial ischemia was significantly decreased(P<0.01),the contents of AST,LDH and CK in serum were significantly decreased(P<0.05,P<0.01),and histopathological examination showed that myocardial injury was significantly alleviated.The protein expression of Bax was significantly decreased(P<0.01),and the protein expressions of Bcl-2,and Bcl-2/Bax ratio were significantly increased(P<0.01).Conclusion Radix Puerariae has a protective effect on myocardial ischemia
