摘 要:目的探讨薏薢方(粉萆薢、薏苡根、威灵仙等)对高尿酸血症模型小鼠、大鼠的降尿酸作用。方法①将60只小鼠随机分为空白组、模型组(次黄嘌呤0.3 g·kg^(-1)+氧嗪酸钾0.05 g·kg^(-1))、苯溴马隆组(0.02 g·kg^(-1))及薏薢高、中、低剂量组(1.33、0.67、0.33 g·kg^(-1)),每组10只;造模的同时连续给药7 d。末次给药后检测小鼠血清尿酸(UA)水平,采用HE染色法观察小鼠肾脏、肝脏组织病理变化。②将60只大鼠随机分为空白组、模型组(次黄嘌呤0.125 g·kg^(-1)+氧嗪酸钾0.05 g·kg^(-1))、苯溴马隆组(0.02 g·kg^(-1))及薏薢高、中、低剂量组(1.92、0.96、0.48 g·kg^(-1)),每组10只;造模的同时连续给药21 d。每周测定大鼠血清UA水平,末次给药后检测大鼠血清肌酐(Cr)、尿素氮(BUN)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL)水平及血清、肝脏黄嘌呤氧化酶(XOD)活性;测定尿UA水平,计算尿酸清除效率;采用HE染色法观察大鼠肾脏、肝脏组织病理变化。结果与空白组比较,模型组大、小鼠的血清UA水平均显著提高(P<0.01)。与模型组比较,薏薢方给药能显著降低高尿酸血症大、小鼠的血清UA水平(P<0.01),改善大、小鼠的肝脏、肾脏组织病理变化;显著降低高尿酸血症大鼠血清Cr、BUN、TG、TC、LDL水平及XOD活性(P<0.05,P<0.01),提高尿UA水平及尿酸清除效率(P<0.05,P<0.01)。结论薏薢方对高尿酸血症模型大鼠、小鼠均具有显著的降尿酸作用,可能与其促进尿酸排泄,抑制血清XOD活性有关。Objective To investigate the uric acid-lowering effect of YiXie Formula(Dioscorea hypoglauca Palibin,Coix lachryma-job;Radix clematidis)on hyperuricemia modeled rats and mice.Methods①Sixty mice were randomly divided into blank group,model group(hypoxanthine 0.3 g·kg^(-1)+potassium oxybenzoate 0.05 g·kg^(-1)),positive drug(benzbromarone)group(0.02 g·kg^(-1))and YiXie Formula high-,medium-and low-dose groups(1.33,0.67,0.33 g·kg^(-1)),10 mice in each group;the drug was administered continuously for 7 days while modeling.The serum uric acid(UA)levels of the mice were measured after the final administration,and the histopathological changes in the kidney and liver of the mice were observed by the HE staining.②Sixty rats were randomly divided into the blank group,the model group(hypoxanthine 0.125 g·kg^(-1)+potassium oxybenzoate 0.05 g·kg^(-1)),positive drug(benzbromarone)group(0.02 g·kg^(-1))and the YiXie Formula high-,medium-and low-dose groups(1.92,0.96 and 0.48 g·kg^(-1)),10 rats in each group;the drug was administered continuously for 21 days while modeling.The serum UA levels of rats were measured weekly,and the serum creatinine(Cr),urea nitrogen(BUN),triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL)levels and serum and liver xanthine oxidase(XOD)activities were measured after the last administration of the drug.Results Compared with the blank group,the serum UA levels of both rats and mice in the model group were significantly increased(P<0.01).Compared with the model group,the YiXie Formula administration groups could significantly reduced the serum UA levels of hyperuricemic rats and mice(P<0.01),and the pathological changes of liver and kidney of rats and mice were improved;the serum Cr,BUN,TG,TC,LDL levels and XOD activity of hyperuricemic rats were significantly reduced(P<0.05,P<0.01),and the urinary UA levels and uric acid clearance rate were improved(P<0.05,P<0.01).Conclusion The YiXie Formula has significant acid-lowering effect on rats and mice with hyperuricemi
