摘 要:胆红素是血红蛋白衰老代谢的主要产物,血液中过量的胆红素可导致脑组织损伤,吸附法是去除血液中过量胆红素的有效方法。本研究以聚苯乙烯大孔吸附树脂为吸附剂载体,依据仿生学原理,采用葡萄糖醛酸对吸附剂进行改性,模仿人体内胆红素与葡萄糖醛酸的结合作用,实现对胆红素的选择性去除。首先对树脂依次进行氯乙酰化、多胺化后,再通过酰胺化反应将葡萄糖醛酸共价键合于树脂上,从而获得胆红素吸附剂。探讨了吸附剂的性能,对吸附时间、温度等吸附参数进行了优化,还研究了人血清白蛋白及离子强度对胆红素吸附的影响。实验结果表明,改性树脂吸附剂对胆红素具有良好的选择性吸附能力,吸附平衡时间为30 min,饱和吸附量为34.5 mg/g,生理浓度范围内的人血清蛋白不影响胆红素的吸附。Bilirubin is the main product of hemoglobin aging metabolism.Excess bilirubin in the blood can cause brain tissue damage.Adsorption is an effective method to remove excess bilirubin in the blood.In this study,polystyrene macroporous adsorbent resin was used as the adsorbent carrier.Based on the principle of bionics,glucuronic acid was used to modify the adsorbent to imitate the binding effect of bilirubin and glucuronic acid in the human body to achieve the selective remove of bilirubin.First,the resin is sequentially chloroacetylated and polyamined,and then glucuronic acid is covalently bonded to the resin through an amidation reaction to obtain a bilirubin adsorbent.The performance of the adsorbent was discussed,and the adsorption parameters such as adsorption time and temperature were optimized.The effects of human serum albumin and ionic strength on the adsorption of bilirubin were also studied.The modified resin adsorbent has good selective adsorption capacity for bilirubin,the adsorption equilibrium time is 30 min,the saturated adsorption capacity is 34.5 mg/g,and the human serum protein within the physiological concentration range does not affect the adsorption of bilirubin.
