摘 要:归纳了目前已报道的磺胺噻唑的8种合成方法。根据起始原料不同,可以分为以对乙酰氨基苯磺酰氯、对硝基苯磺酰氯、对乙酰氨基磺胺硫脲和对硝基苯二硫醚为起始原料4类。并且结合每条合成工艺路线,讨论和现有合成方法的优缺点。基于现有研究,以对乙酰氨基苯磺酰氯为原料和2-氨基噻唑反应具有高收率、生产操作简单的特点,是一条适合于工业化生产的合成路线。认为寻找适宜的缚酸剂,扩大溶剂体系研究,选择更加绿色安全的溶剂体系是未来研究的方向。Eight synthetic methods of sulfathiazole were summarized. According to different starting materials, they can be divided into four kinds of starting materials: p-acetylaminobenzenesulfonyl chloride, p-nitrobenzenesulfonyl chloride, pacetylaminosulfonamide thiourea and p-nitrobenzene disulfide. In addition, the advantages and disadvantages of the existing synthetic methods were discussed in combination with each synthetic route. Based on the existing research, the reaction of 2-aminothiazole with p-acetylaminosulfonyl chloride as raw material has the characteristics of high yield and simple operation, which is a suitable synthetic route for industrial production. It is considered that the future research direction is to find suitable acid-binding agent, expand the research of solvent system and select more green and safe solvent system.
