摘  要:为了揭示川西高海拔地区(四川甘孜藏族自治州和阿坝藏族羌族自治州)原生野生饲用黄芪根瘤菌的遗传多样性,为筛选优良菌株提供依据,采用纯培养法从该地区部分饲用黄芪植物根瘤中分离纯化根瘤菌;通过BOXAIR、16S rDNA-RFLP及PCA(principal component analysis)分析饲用黄芪根瘤菌的遗传多样性。结果表明,从川西高海拔地区7个县15个采样点获得10种饲用黄芪植物的根瘤,共分离纯化出36个根瘤菌菌株;在相似系数82.4%的16SrDNAPCR-RFLP分析中,所有供试菌株均与参比菌株完全分开,供试菌株聚成8个遗传群(SCAU509,SCAU553单独分开);在相似系数83.2%的BOX-PCR分析中,饲用黄芪根瘤菌36个供试菌株和14个参比菌株聚成10个遗传群。饲用黄芪根瘤菌16S rDNA Simpson遗传多样性指数D=0.949。研究显示川西高海拔地区(四川甘孜藏族自治州和阿坝藏族羌族自治州)饲用黄芪根瘤菌具有丰富的遗传多样性。To provide a basis for screening excellent strains, the genetic diversity of the rhizobial bacteria nodulating fodder plants of Astragalus in the high altitude area of the western Sichuan(Ganzi Tibetan Autonomous Prefecture and Aba Tibetan Qiang Autonomous Prefecture, Sichuan, China) were evaluated. The pure culture method was used for isolating the rhizobial strains from the nodules;BOXAIR, 16S rDNA PCR-RFLP and principal component analysis(PCA) were used for the determination of the genetic diversity. As results, thirty-six strains were isolated from fodder plants of Astragalus in 15 samples from 7 counties in the western Sichuan. The tested strains were classified into 8 clusters at similarity of 82.4% in 16S rDNA PCR-RFLP, except that SCAU509and SCAU553 were separated, and 10 clusters at similarity of 83.2% were formed in BOXAIR. The 16S rDNA Simpson genetic diversity index was D = 0.949. This study revealed the high genetic diversity of the rhizobia isolated from fodder plants of Astragalus in high altitude area of the western Sichuan.
