摘 要:本实验主要从代谢组出发,构建浓香型烟草(Nicotiana tabacum)品种‘NC89’叶片在不同氮素水平下的差异代谢物数据矩阵,通过聚类分析和代谢通路富集分析来确定‘NC89’的代谢组特色和适宜肥力,为进一步提高其烟叶质量提供理论依据。结果表明,随着施氮量增加,‘NC89’叶片中含量降低的代谢物数量整体呈增加趋势;而且在高氮(≥45 kg·hm^(-2))和低氮(≤37.5 kg·hm^(-2))条件下,‘NC89’叶片的代谢模式是不同的,其中苯丙氨酸代谢以及丙氨酸、天冬氨酸和谷氨酸代谢是受施氮量影响最大的两条代谢通路。分析两个代谢通路中差异代谢物,发现随着施氮量增加,烟叶中与香气物质和次生代谢物有关的苯丙氨酸含量先升高后持续降低,与逆境胁迫相关的γ-氨基丁酸含量先升高后降低再升高,烟叶的碳氮代谢逐渐失衡。考虑烟叶碳氮代谢平衡、烟叶外观与香气质量、经济与环境效益等方面,认为烟草品种‘NC89’在许昌市襄城县烟区的适宜施氮量为37.5 kg·hm^(-2)。In this experiment, the data matrix of differential metabolites in leaves of strong flavor type tobacco(Nicotiana Tabacum) variety ’NC89’ under different nitrogen levels was constructed based on the metabolome, and the metabolomic characteristics and suitable fertility of ’NC89’ were determined by cluster analysis and metabolic pathway enrichment analysis, so as to provide a theoretical basis for further improving the quality of tobacco leaves. The results show that with the increase of nitrogen application rate, the number of metabolites with decreased content in ’NC89’ leaves showed an overall increasing trend. Moreover, metabolic patterns of ’NC89’ were different under the conditions of high nitrogen(≥45kg·hm^(-2)) and low nitrogen(≤37.5 kg·hm^(-2)). Besides, phenylalanine metabolism and alanine, aspartic acid and glutamic acid metabolism were the two most affected metabolic pathways by nitrogen application rate. The analysis of differential metabolites in these two metabolic pathways showed that with the increase of nitrogen application rate, the content of phenylalanine related to aroma substances and secondary metabolites in tobacco leaves increased at first and then decreased continuously;the content of γ-aminobutyric acid related to stress increased at first, then decreased and then increased;the carbon metabolism and the nitrogen metabolism of tobacco leaves were gradually out of balance. Based on the consideration of the balance of carbon and nitrogen metabolism, the quality of appearance and aroma of tobacco leaves, economic and environmental benefits, it is concluded that the suitable nitrogen application rate of tobacco variety ’NC89’ is 37.5 kg·hm^(-2)in tobacco area of Xiangcheng County of Xuchang City.
