摘 要:目的探讨葡萄内酯抗血小板活化作用及其分子机制。方法制备洗涤血小板和富含血小板血浆,仪器检测胶原、凝血酶、ADP、花生四烯酸等激动剂对血小板聚集的影响;流式细胞仪检测血小板CD62P和JON/A表达;Western blot检测IκBα,PLCγ2,Syk等蛋白的磷酸化水平;采用FeCl_(3)诱导的小鼠颈动脉血栓模型评价葡萄内酯对体内动脉血栓形成的影响。结果葡萄内酯可有效抑制血小板聚集,且对胶原诱导的血小板聚集作用抑制明显(P<0.05);葡萄内酯可显著抑制胶原刺激引起的血小板释放和CD62P、JON/A表达(P<0.05)和降低IκBα、PLCγ2、Syk等蛋白的磷酸化水平(P<0.05),且可明显延长Fe Cl3诱导的小鼠颈动脉血流闭合时间(P<0.05)。结论葡萄内酯可通过NF-κB介导的PLCγ2-Syk信号通路发挥抗血小板作用以及抑制动脉血栓形成。Objective To investigate the antiplatelet activation and its molecular mechanism of auraptene.Methods Washing platelets and platelet-rich plasma were prepared,and the effects of agonists such as collagen,thrombin,ADP and arachidonic acid on platelet aggregation were detected.The expression of CD62P and JON/A on platelets were detected by flow cytometry.The phosphorylation levels of IκBα,PLCγ2 and SYK were detected by Western blot.The effect of aurapten on the thrombosis of mice carotid artery induced by Fe Cl3 was evaluated.Results Auraptene could effectively inhibit platelet aggregation induced by collagen(P<0.05).Aurapten also inhibited the expression of CD62p、JON/A on membrane(P<0.05),and decreased the phosphorylation levels of IκBα、PLCγ2 and SYK(P<0.05),and significantly prolonged the closure time of carotid artery blood flow in mice(P<0.05).Conclusion Auraptene can play an antiplatelet role and inhibit arterial thrombosis through PLCγ2-SYK signaling pathway mediated by NF-κB.
