摘 要:目的 从BDNF信号通路角度探讨红景天胶囊的抗抑郁作用机制。方法 采用慢性温和不可预知应激(CUMS)程序对大鼠进行造模,以高、低剂量红景天胶囊及舍曲林为干预药物,检测各组大鼠糖水偏爱率、旷场实验、强迫游泳实验、血清脑源性神经生长因子(BDNF)、环磷酸腺苷反应原件结合蛋白(CREB)、糖原合成酶激酶-3β(GSK-3β)水平及海马组织中CREB、BDNF、GSK-3β mRNA水平。结果 与对照组相比,模型组的糖水偏爱率、旷场实验、血清CREB、BDNF及海马组织中的CREB、BDNF mRNA水平均显著下降,强迫游泳不动时间、血清GSK-3β及GSK-3β mRNA水平升高(P<0.05),红景天胶囊干预后,与模型组对比,各剂量组糖水偏爱率、旷场实验、血清CREB、BDNF及海马组织中的CREB、BDNF mRNA水平均出现回升,强迫游泳不动时间、血清GSK-3β及GSK-3β mRNA水平下调(P<0.05)。结论 红景天胶囊可通过调控BDNF/GSK-3β/CREB信号通路发挥抗抑郁的作用。Objective To investigate the anti-depressant mechanism of Rhodiola capsule from the perspective of BDNF signaling pathway.Methods The rats’ model was established by chronic unpredictable mild stress(CUMS). The rats were administrated with high and low doses of Rhodiola capsule and sertraline as intervention drugs. The levels of sucrose preference test, open-field test, forced swimming test,BDNF, CREB and GSK-3β in serum, protein expression of BDNF, CREB and GSK-3β in tissues and their corresponding mRNA levels were explored for their antidepressant mechanism.Results Compared with the blank group, the levels of sucrose preference test, open-field test,CREB and BDNF in serum, expression of CREB and BDNF in the hippocampus of the model group were significantly decreased. But the dead time of forced swimming test, the expression of GSK-3β in serum and GSK-3β mRNA were significantly increased(P< 0. 05,P<0. 01). After the intervention of Rhodiola capsule, compared with the model group, there was a significant difference in the callus of each dose group. At the same time, the levels of sucrose preference test, open-field test, CREB and BDNF in serum, the mRNA of CREB and BDNF in the hippocampus of the model group were significantly increased. And the dead time of forced swimming test, the expression of GSK-3β in serum and GSK-3β mRNA were significantly decreased(P< 0. 05,P< 0. 01). After the intervention, there was a callback in each dose group compared with the model group, and there had significant differences.Conclusion Rhodiola capsule can exert antidepressant effect by regulating BDNF/GSK-3β/CREB signaling pathway.
