摘  要:目的合成一种新型FeGd纳米复合材料作为T_(1)-T_(2)双模态纳米磁共振(MRI)造影剂,并在体外探索其双模态的造影性能以及生物安全性。方法利用透射电子显微镜(TEM)观察FeGd纳米复合材料的形貌,通过纳米粒度仪分析FeGd纳米复合物的水合粒径,借助傅里叶变换红外光谱仪和热重分析仪探索FeGd纳米复合物的组分,通过7.0 T小动物磁共振成像仪分析FeGd纳米复合物的双模态磁共振造影性能,利用CCK-8实验评价FeGd纳米复合物的细胞生物安全性。结果制备的FeGd纳米复合材料的尺寸约为150 nm,呈现球形,表面电位为正电。纳米复合材料具有良好的胶体稳定性,且其组分中含有17.6%的聚乙烯吡咯烷酮有机物。FeGd纳米复合材料的r_(1)和r_(2)值分别为6.765 mM^(-1)·s^(-1)和213.81 mM^(-1)·s^(-1),这证明了FeGd造影剂具有很好的双模成像性能。FeGd纳米复合材料对正常细胞C166和血红细胞几乎没有损伤,展现出良好的生物相容性。结论新型的FeGd纳米复合物具有良好的T_(1)和T_(2)双模态纳米磁共振造影性能,展现出良好的应用前景。Objective To synthesize a novel FeGd nanocomposite as T_(1)-T_(2) dual-mode magnetic resonance imaging(MRI)contrast agent,and explore its dual-mode contrast performance and biosafety in vitro.Methods The morphology of FeGd nanocomposites was observed through transmission electron microscope(TEM).The hydrodynamic size of FeGd nanocomposites was analyzed through a dynamic laser scattering device.The composition of FeGd nanocomposites were explored through fourier transform infrared spectra(FT-IR)and thermal gravimetric(TG)analysis.MRI contrast performance of FeGd nanocomposites was investigated through 7.0 T MRI scanner.CCK-8 assay was used to evaluate the cell biosafety of FeGd nanocomposites.Results The prepared FeGd nanocomposites showed spherical shape with about 150 nm and possessed excellent colloidal stability.Moreover,the surface of FeGd nanocomposites presented positive charge.The composition of FeGd composites contained approximate 17.6%of polyvinylpyrrolidone(PVP)polymer.MRI results indicated that the r_(1) and r_(2) values of FeGd composites were 6.765 mM^(-1)·s^(-1) and 213.81 mM^(-1)·s^(-1),respectively,confirming excellent T_(1)-T_(2) dual-modal contrast performance of FeGd composites.In terms of safety,FeGd composites had no any damage on normal cells and red blood cells,showing excellent biosafety.Conclusion A novel FeGd nanocomposite has excellent T_(1)-T_(2) dual-modal MRI contrast performance,showing the great application prospect.
