摘 要:目的利用生物信息学分析亚急性皮肤型狼疮(SCLE)的差异表达基因(DEGs)、关键信号通路及免疫细胞浸润情况,为SCLE的认识、药物开发等提供新思路。方法筛选GSE81071和GSE109248中SCLE和正常对照(NC)皮肤活检组织DEGs,对其进行基因本体论(GO)分析、京都基因与基因组百科全书(KEGG)通路富集分析,采用蛋白质-蛋白质相互作用(PPI)网络及关键分子筛选识别核心基因,利用CIBERSORT分析22种免疫细胞浸润。结果筛选出34个DEGs,生物学过程主要集中在对病毒的防御反应、Ⅰ型干扰素信号通路等。分子功能主要包括双链RNA结合、腺苷酸转移酶活性、CXCR趋化因子受体结合等。PPI网络分析筛选出前3个核心基因:CXCL10、DDX58和RSAD2。与NC组相比,SCLE组CD4+活化记忆T淋巴细胞比例明显升高(P=0.007),而Tregs比例明显降低(P=0.001)。结论CXCL10、DDX58和RSAD2可能是预测SCLE的生物学标志物及潜在的治疗靶点。Objective To analyze the differentially expressed genes(DEGs),key signal pathways and immune cell infiltration in subacute cutaneous lupus erythematosus(SCLE)by bioinformatics,so as to provide new ideas for the understanding and drug development of SCLE.Methods The DEGs of SCLE and normal control(NC)skin biopsies from GSE81071 and GSE109248 were selected,and the gene ontology(GO)analysis and enrichment analysis of Kyoto Encyclopedia of Genes and Genomes(KEGG)were carried out.The core genes were identified by protein-protein interaction(PPI)network and key molecular screening,and the infiltration of 22 kinds of immune cells was analyzed by CIBERSORT.Results A total of 34 DEGs were screened out,and the biological process mainly focused on the defense response to viruses and the typeⅠinterferon signaling pathway,etc.Molecular functions mainly include double-stranded RNA binding,adenylyl transferase activity,and CXCR chemokine receptor binding,etc.The top three core genes,CXCL10,DDX58 and RSAD2,were screened by PPI analysis.Compared with the NC group,the percentage of CD4+activated memory T lymphocytes in the SCLE group was increased significantly(P=0.007),while the percentage of Tregs was decreased significantly(P=0.001).Conclusion CXCL10,DDX58 and RSAD2 might be predictive biomarkers and potential therapeutic target for SCLE.
