摘 要:目的 研究茺蔚子Leonuri Fructus化学成分及其肝细胞保护作用。方法 对茺蔚子正丁醇提取物采用大孔吸附树脂、HW-40F凝胶以及半制备液相色谱进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用对乙酰氨基酚(APAP)所致HL-7702细胞损伤模型评价其肝细胞保护作用。结果 从中分离得到7个苷类,分别鉴定为β-苯乙醇葡萄糖苷(1)、darendoside B(2)、肉苁蓉苷E(3)、肉苁蓉苷G(4)、熊果苷(5)、2-O-β-葡萄糖基-5-甲氧基-苯甲酸甲酯(6)、对羟基苯甲酰基熊果苷(7)。其中化合物4、5具有肝细胞保护作用,其EC_(50)值分别为(35.71±6.26)、(6.66±0.79)μmol/L。结论 化合物2、4~7均为首次从该植物中分离得到,化合物4、5均具有显著的肝细胞保护作用。AIM To study the chemical constituents from Leonuri Fructus and their hepatocellular protective effect.METHODS The n-butanol extract from Leonuri Fructus was isolated and purified by macroporous adsorption resin and HW-40 F column chromatography and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their protective effects activities on hepatocytes were evaluated by acetaminophen(APAP)-induced HL-7702 cell injury model.RESULTS Seven compounds were isolated and identified as β-phenylethanol glucoside(1),darendoside B(2),cistanoside E(3),cistanoside G(4),arbutin(5),2-O-β-glucopyranosyl-5-methoxy-benzoic acid methyl ester(6),lanceoloside A(7).Compounds 4 and 5 exhibited protective effects with EC_(50) values of(35.71±6.26) and(6.66±0.79) μmol/L,respectively.CONCLUSION Compounds 2 and 4-7 are isolated from this plant for the first time.Compounds 4 and 5 show significant hepatoprotective effects.
