摘  要:为了对牛坏死杆菌43K OMP进行生物信息学分析,并预测其B细胞表位,研究利用Prot Param在线软件、TMHMM在线软件、Net Phos软件、DNA star软件和I-TASSER在线软件,分别对43K OMP的理化性质、跨膜区、磷酸化位点、二级结构和三级结构进行分析,用Bepipred在线软件和Ellipro在线软件对其B细胞表位进行预测,并对预测的表位肽进行验证。结果表明43K OMP共编码377个氨基酸,相对分子质量为42.927×10^(3),理论等电点为8.74,该蛋白在1-20aa处存在信号肽区域,无跨膜区,包括34个磷酸化位点,具有10个构象B细胞表位,多个线性B细胞表位,合成的4表位肽经ELISA验证显示反应性良好。通过生物信息学技术分析出43K OMP的性质及结构,预测出其具有多个B细胞表位,为该蛋白的功能研究及基因工程疫苗的研制提供依据。In order to analyze the 43K OMP of bovine Fusobacterium necrophorum in bioinformatics and predict its B cell epitope,it used Prot Param online software,TMHMM online software,Net Phos software,DNA star software and I-TASSER online software to analyze the 43K Analyze the physical and chemical properties,transmembrane region,phosphorylation site,secondary structure and tertiary structure of OMP,use Bepipred online software and Ellipro online software to predict its B cell epitope,and verify the predicted epitope peptide.The results showed that 43K OMP encodes a total of 377 amino acids,the relative molecular mass was 42.927×10^(3),and the theoretical isoelectric point was 8.74.The protein had a signal peptide region at 1-20 aa,no transmembrane region,and included 34 phosphorylation sites.It had 10 conformational B cell epitopes and multiple linear B cell epitopes.The synthesized 4-epitope peptide had been verified by ELISA and showed good reactivity.The nature and structure of 43K OMP were analyzed through bioinformatics technology,and it was predicted to have multiple B cell epitopes,which provided a basis for the functional study of the protein and the development of genetic engineering vaccines.
