摘 要:目的 探究养阴通脑颗粒对脑缺血再灌注损伤大鼠的保护作用。方法 建立大鼠大脑中动脉栓塞(MCAO)模型,SD大鼠随机分为假手术组、模型组、养阴通脑颗粒组。建立模型后24、48、72 h分别测定神经功能评分,术后72 h进行TTC染色并计算脑梗死体积,逆转录-聚合酶链式反应法(RT-PCR)检测大鼠脑组织中NLRP3炎性小体、信号传导与转录激活因子(STAT3) mRNA表达,酶联免疫吸附法检测血浆中白细胞介素-18(IL-18)、肿瘤坏死因子-α(TNF-α)水平。结果 养阴通脑颗粒能改善神经功能,减少脑梗死体积,显著降低IL-18和TNF-α含量,下调NLRP3、STAT3 m RNA表达。结论 养阴通脑颗粒对大鼠脑缺血再灌注损伤有一定的保护作用,其作用机制可能与抑制炎症反应有关。Objective:To investigate the protective effect of Yangyin Tongnao Granules on cerebral ischemia/reperfusion injury in rats.Methods:The middle cerebral artery occlusion(MCAO)model was established in rats.SD rats were randomly divided into sham operation group,model group and Yangyin Tongnao Granules group.Neurological function scores were measured at 24,48,and 72 hours after the model was established.TTC staining was performed at 72 hours after operation to calculate the volume of cerebral infarction.Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect NLRP3 inflammation and mRNA expression of signal transduction and activator of transcription(STAT3)in rat′s brain tissue,and the levels of interleukin-18(IL-18)and tumor necrosis factor-α(TNF-α)in plasma were detected by enzyme-linked immunosorbent assay.Results:Yangyin Tongnao Granules could improve the neurological function,reduce the volume of cerebral infarction,significantly reduce the contents of IL-18 and TNF-α,and down-regulate the mRNA expression of NLRP3 and STAT3.Conclusion:Yangyin Tongnao Granules has a certain protective effect on cerebral ischemia-reperfusion injury in rats,and its mechanism may be related to inflammatory reaction.
