摘 要:目的 观察中药补肺颗粒含药血清对哮喘小鼠支气管平滑肌细胞(ASMC)增殖以及气道血管生成的影响。方法 制备补肺颗粒含药血清,参照文献制备哮喘小鼠模型并分离ASMC,随机分为对照组、高剂量组和低剂量组,分别给予不含药血清、高剂量含药血清及低剂量含药血清加以培养,后测定ASMC增殖率,Western blotting测定ASMC血管内皮生长因子(VEGF)及内皮抑素(ES)的相对表达,RT-PCR检测ASMC VEGF mRNA及ES mRNA相对表达。结果 与对照组对比,高剂量组和低剂量组能显著降低ASMC增殖(P <0.01),VEGF及内皮抑素的相对表达显著降低(P <0.05),高剂量组的VEGF m RNA及ES mRNA的相对表达显著降低(P <0.01)。结论 补肺颗粒含药血清能有效抑制哮喘小鼠气道平滑肌细胞增殖,进而改善哮喘气道重塑状态。其作用机制可能与抑制VEGF及内皮抑素表达,调节气道血管生成有关。Objective:To observe the effect of drug containing serum of Bufei Granules on the proliferation of bronchial smooth muscle cells(ASMC)and airway angiogenesis in asthmatic mouse.Methods:Drug containing serum of Bufei Granules was prepared.Asthmatic mouse model was prepared according to literature and ASMC was isolated and randomly divided into control group,high-dose group and low-dose group.They were cultured with drug-free serum,high-dose drug-containing serum and low-dose drug-containing serum separately.The proliferations of BSMC and the relative expression of VEGF mRNA and ES m RNA in ASMC were detected.Results:Compared with the control group,the proliferation of ASMC in high dose group and low dose group was significantly decreased(P < 0.01),the relative expression of VEGF and endostatin was significantly decreased(P < 0.05),the relative expression of VEGF mRNA and ES m RNA in high dose group was significantly decreased(P < 0.01).Conclusion:Drug containing serum of Bufei Granules can effectively inhibit the proliferation of airway smooth muscle cells in asthmatic mice,and inhibit the airway remodeling.The mechanism of the action may be related to the inhibition of VEGF and endostatin expression and the regulation of airway angiogenesis.
