摘 要:端粒是真核生物染色体物理末端的非转录重复序列(TTAGGG)n及一些结合蛋白组成的特殊结构,在染色体复制、末端保护和细胞寿命等方面具有重要作用。TRB(telomere repeat binding)是一类端粒双链DNA结合蛋白,其关键DNA结合结构域位于N端的Myb结构域。TRB蛋白可以与端粒相关蛋白POT1b(protection of telomeres 1b)、TERT(telomerase reverse transcriptase)等互作,具有调节端粒长度、募集相关因子、影响端粒酶活性等端粒功能。TRB蛋白还可以与非端粒蛋白CHR19(chromatin remodeling protein 19)、HDT4(histone deacetylase 4)、HDA6(histone deacetylase 6)、CLF(curly leaf)、SWN(swinger)等互作,TRB通过与telobox相关基序结合参与转录调控,具有非端粒功能。本文介绍了植物TRB蛋白的主要结构,概述了其端粒与非端粒功能,构建了TRB蛋白功能网络,为丰富TRB蛋白多功能网络机制研究提供了参考。Telomere is a special structure composed of non-transcriptional repeat sequences(TTAGGG)_(n)and some telomeric binding proteins at the physical end of eukaryotic chromosomes.It plays important roles in chromosome replication,protection and cell life,etc.TRB(telomere repeat binding)proteins are kind of telomere DNA double-stranded binding proteins.Its key region is the Myb domain at the N-terminu.TRB proteins could bind to telomere double-stranded DNA,as well as interact with telomere related proteins,such as POT1b(protection of telomeres 1b)and TERT(telomerase reverse transcriptase),which might regulate telomere length,recruite to telomeres and affect telomerase acticity.Recently,TRB proteins could also interact with non-telomeric proteins,including CHR19(chromatin remodeling protein 19),HDT4(histone deacetylase4),HDA6(histone deacetylase 6),CLF(curry leaf),SWN(swinger),which may have non-telomeric function involved in transcriptional regulation by binding with telobox related motifs.This paper summarizd the main structure of plant TRB proteins,recommended the research progress on telomeric and non-telomeric function,constructed the TRB protein functional network,which could provide a reference for enriching the construction of TRB proteins multifunctional network mechanism.
