摘  要:目的探讨人工脑脊液对于原代培养的皮层神经元和小脑颗粒细胞的功能维持作用及对损伤神经元的保护作用。方法制备人工脑脊液,并初步考查其稳定性。以氯化钠注射液、乳酸钠林格注射液及醋酸钠林格注射液为阳性对照,采用MTT比色法检测药物对皮层神经元和小脑颗粒细胞存活率的影响,评价不同药物对谷氨酸、过氧化氢及机械划伤所致损伤神经元的保护作用。结果自制人工脑脊液在加速试验条件下稳定性良好。与各阳性对照组比较,其在维持皮层神经元和小脑颗粒细胞存活率方面的效果更佳,对损伤神经元具有更好的保护作用。当人工脑脊液处方中缺乏钙、镁或磷元素时,均会降低细胞活性。结论自制人工脑脊液质量稳定,具有维持神经细胞正常生理功能与保护损伤细胞的作用。OBJECTIVE To investigate the functional maintenance effect of artificial cerebrospinal fluid(ACSF)on primary cultured cortical neurons and cerebellar granule cells and its protective effect on injured neurons.METHODS The ACSF was prepared and the stability trial for this production was carried out.With sodium chloride injection,sodium lactate Ringer’s injection,and sodium acetate Ringer’s injection as positive controls,MTT assay was used to evaluate the functional maintenance effect of ACSF on primary cultured cortical neurons and cerebellar granule cells and the neuroprotective effect on injured neurons caused by glutamate,hydrogen peroxide,or mechanical scratches separately.RESULTS The self-made ACSF had good stability under accelerated conditions.It maintained the function of cortical neurons and cerebellar granule cells and showed the neuroprotective effect on the injured neurons better than the positive controls.The absence of Ca,Mg,or P in the formulation of ACSF would decrease the cell activity.CONCLUSION The self-made ACSF is of stable quality and has the neuroprotective effect of maintaining the normal physiological functions of nerve cells and reducing injury.
