摘 要:合成了含吡啶环的2-氨基-5-硝基吡啶水杨醛Schiff碱(HL),产物经红外光谱、质谱、核磁共振波谱进行了确认。通过电子光谱法和分子发光光谱法研究了HL与鲱鱼精DNA(hs-DNA)的相互作用。结果表明:HL可引起hs-DNA 260 nm处特征吸收峰的红移和明显的减色效应,HL可竞争溴化乙锭(EB)与DNA的结合,把与hs-DNA结合的EB释放出来,令EB-DNA体系发生静态荧光猝灭。热力学计算表明,氢键和范德华力是两者间主要作用力类型,HL以嵌插方式与hs-DNA结合。2-amino-5-nitropyridine salicylaldehyde Schiff base(HL)containing pyridine ring was synthesized.The product was confirmed by IR spectrum,Mass spectrometry and NMR spectroscopy.The interaction between HL and herring sperm DNA(hs-DNA)was studied by electron spectroscopy and molecular luminescence spectroscopy.The results showed that,HL could cause red shift of hs-DNA’s characteristic absorption peak at 260nm and obvious color subtraction effect.HL competed binding of ethidium bromide(EB)to hs-DNA.The fluorescence of EB-hs-DNA system was significantly quenched,and the quenching type was static quenching.Thermodynamic studies showed that,the interaction force between complexes was mainly hydrogen bond and van der Waals force,and HL binded to hs-DNA by intercalation.
