摘 要:为了解上海郊区猪场戊型肝炎病毒(HEV)的感染情况,从3个猪场随机采集180份新鲜粪便样本,用逆转录巢式聚合酶链反应对HEV ORF2部分基因片段进行扩增、克隆及测序。结果表明:HEV RNA的PCR检测总阳性率为15.6%,个别猪场HEV RNA检出率达30%,且存在基因3型和4型同时检出的情况。经NCBI数据库BLAST比对分析,23份样本为基因4型,5份样本为基因3型。遗传进化分析显示,基因3型和4型毒株与来源于上海地区的参考毒株遗传关系最近,核苷酸同源性分别为97.9%和96.3%。研究表明,上海郊区某些猪场HEV感染率仍较高,且存在基因3型与4型共感染的现象,应注意加强防控。In order to understand the infection of hepatitis E virus(HEV)in pig farms in Shanghai suburb,a total of 180 fresh fecal samples were randomly collected from 3 pig farms,and partial fragment of HEV ORF2 gene were amplified,cloned and sequenced by reverse transcription nested polymerase chain reaction.The results showed that the overall positive rate of HEV RNA detected by PCR was 15.6%,and the detection rate of HEV RNA in individual pig farms was 30%,and genotype 3 and genotype 4 were detected at the same time.Through the BLAST comparative analysis of NCBI database,23 samples were classified into genotype 4 and 5 samples were classified into genotype 3.Phylogenetic analysis showed that the strains of genotype 3 and genotype 4 had the closest genetic relationship with the reference strains from Shanghai,with nucleotide homology of 97.9%and 96.3%respectively.The infection rate of HEV in some pig farms in suburb of Shanghai was still relatively high,and there was the phenomenon of coinfection of genotype 3 and genotype 4.Attention should be paid to strengthening prevention and control.
