摘 要:为研究小麦(Triticum aestivum)GLR(Glutamate receptor)基因家族的特征,通过生物信息学的方法分析了其理化性质,系统进化,染色体位置,基因结构,保守结构域及启动子顺式作用元件。利用RT-qPCR技术分析TaGLR基因组织表达及低温、盐、热和干旱胁迫下的表达变化。结果表明,小麦全基因组中鉴定到100个GLR基因序列,分布于小麦的17条染色体上。系统进化分析表明TaGLR可分为4个亚家族,结构域分析发现,GLR编码蛋白结构保守,均包含两个谷氨酸受体结合区(GlnH1和GlnH_(2))和4个跨膜区域(M1,M2,M3,M4)。TaGLR基因家族成员启动子区域均包含响应植物激素和逆境胁迫的顺式调控元件。组织表达分析结果表明,TaGLR1.1-A.1、TaGLR2.3-A、TaGLR3.4-B、TaGLR3.9-A在各组织中均有表达,其他成员基本不表达。TaGLR2.3-A、TaGLR3.4-B、TaGLR3.9-A表达受逆境胁迫影响,TaGLR2.3-A在干旱胁迫下表达变化极显著(在耐旱品种中表达显著增加,在敏感品种中表达显著降低),逆境处理条件下TaGLR3.4-B、TaGLR3.9-A基因表达均受到抑制。本研究首次鉴定小麦GLR基因家族并对其结构和功能进行初步解析,为深入研究GLR基因与逆境胁迫的关系提供了理论基础。In order to study the characteristics of GLR(Glutamate receptor)gene family of wheat(Triticum aestivum),bioinformatics methods were used to analyze its physical and chemical properties,phylogeny,chromosome location,gene structure,conserved domains and cis-acting of promoters.RT-qPCR technique was used to analyze the tissue expression of TaGLR genes and their expression changes under low temperature,salt,heat and drought stress.The results showed that a total of 100 GLR gene sequences were identified in the wheat genome,which were distributed on 17 chromosomes of wheat.Phylogenetic analysis showed that TaGLR genes could be divided into 4 subfamilies,which were both different and conservative in evolution.According to domain analysis,the GLR structure of other plants is similar to its encoded protein structure,and they all contain two Glu receptor binding regions(GlnH1 and GlnH_(2)),and 4 transmembrane regions(M1,M2,M3,M4).The promoter regions of TaGLR gene family was found to have cis regulatory elements related to plant hormone and stress response.Tissue expression analysis results show that TaGLR1.1-A.1,Ta GLR2.3-A,TaGLR3.4-B and TaGLR3.9-A were expressed in all tissues,and the other members basically did not express.The expression of TaGLR2.3-A,TaGLR3.4-B,and TaGLR3.9-A were all affected by adversity stress,and TaGLR2.3-A mainly responded to drought stress(the expression increased significantly in drought tolerant varieties and decreased significantly in sensitive varieties),and the expression of TaGLR3.4-B and TaGLR3.9-A genes were inhibited under stress treatment conditions.This study identified the wheat GLR gene family for the first time and preliminarily analyzed its structure and function,which provided a theoretical basis for further study on the relationship between GLR gene and adversity stress.
