摘 要:为优化小花矮牵牛(Calibrachoa hybrida)茎尖和茎段外植体的离体繁殖技术。本研究采用的实验材料为小花矮牵牛(Calibrachoa hybrid’lindura yellow’),取茎尖和茎段作为外植体。首先对外植体进行原生质体的分离和纯化,然后接种在添加不同浓度的植物生长调节剂的MS培养基上进行愈伤组织的诱导、分化、增殖和生根。植物生长调节剂包括3-吲哚丁酸(IBA)、细胞分裂素(BAP)、萘乙酸(NAA)、二氯苯氧乙酸(2,4-D)和激动素(Kinetin)。结果显示,IBA(1.0 mg/L)+BAP(1.0 mg/L)生长调节剂组合下获得的茎尖(95.63%)和茎段(92.74%)微愈伤组织形成率最高。BAP(1.5 mg/L)+2,4-D(1.5 mg/L)生长调节剂组合下获得的茎尖(95.08%)和茎段(94.31%)愈伤组织诱导率最高。Kinetin(2.0 mg/L)+NAA(1.5 mg/L)生长调节剂组合下获得的茎尖(37.53%)和茎段(29.54%)再生率最高,且茎尖(15.63)和茎段(12.27)的出芽数最高。BAP(1.0 mg/L)+IBA(1.0 mg/L)生长调节剂组合下获得的茎尖(94.55%)和茎段(91.53%)芽增殖率最高,茎尖(18.06)和茎段(16.34)的芽数最高,茎尖(2.33 cm)和茎段(2.15 cm)的芽长最高。IBA(1.5 mg/L)处理下获得的茎尖(8.24)和茎段(8.05)根数最高。总之,本研究优化了以小花矮牵牛茎尖和茎段为外植体的离体繁殖方案,确定了适合小花矮牵牛愈伤组织的诱导、分化、增殖和生根的最佳培养基。In order to opti mize the in vitro propagation technique of shoot tip and stem segment explants of Calibrachoa hybrida.In this study,the experimental material was Calibrachoa hybrid’lindura yellow’,and the shoot tips and stem segments were taken as explants.Protoplasts were first isolated and purified from explants,then inoculated on MS medium supplemented with various concentrations of plant growth regulators for callus induction,differentiation,proliferation and rooting.Plant growth regulators include 3-indolebutyric acid(IBA),cytokinin(BAP),naphthalene acetic acid(NAA),dichlorophenoxyacetic acid(2,4-D)and kinetin.The results showed that the microcallus formation rate of shoot tips(95.63%)and stem segments(92.74%)obtained under the combination of IBA(1.0 mg/L)+BAP(1.0 mg/L)growth regulator was the highest.The callus induction rate of shoot tips(95.08%)and stem segments(94.31%)obtained under the combination of BAP(1.5 mg/L)+2,4-D(1.5 mg/L)growth regulator was the highest.The regeneration rate of shoot tips(37.53%)and stem segments(29.54%)obtained under the combination of Kinetin(2.0 mg/L)+NAA(1.5 mg/L)growth regulator was the highest,and the shoot tips(15.63)and stem segments(12.27)had the highest number of buds.The shoot tip(94.55%)and stem segment(91.53%)obtained under the combination of BAP(1.0 mg/L)+IBA(1.0 mg/L)growth regulator had the highest bud proliferation rate,and the shoot tip(18.06)and stem segment(16.34)had the highest bud number,and the shoot tip(2.33 cm)and stem segment(2.15 cm)had the highest bud length.The number of shoot tips(8.24)and stem segments(8.05)obtained by IBA(1.5 mg/L)treatment was the highest.In conclusion,this study optimized the in vitro propagation scheme using the shoot tips and stem segments of Calibrachoa hybrida as explants,and determined the best medium suitable for the induction,differentiation,proliferation and rooting of Calibrachoa hybrida.
