摘  要:以珍稀濒危树种河北梨(Pyrus hopeiensis Yu)的实生无菌苗为材料,通过茎尖和芽尖组织培养,比较了不同激素配比对河北梨组培繁殖能力的影响。结果表明,河北梨“三低”现象(结实率低,种子发芽率低,枝条生根率低)是河北梨种群扩散困难的主要原因,实生苗的茎芽组培繁殖是有可能解决河北梨种群扩繁难题的重要途径;不同激素配比的培养条件下,基础培养基为1/2MS,添加6-BA(0.5 mg/L)、NAA(0.05 mg/L)和TDZ(0.2 mg/L)是最佳的继代培养基组成,扩繁系数可达到3.91。本研究建立了珍稀濒危植物河北梨的组织培养快速繁殖技术体系,为该树种的迁地保护提供理论依据和技术规程。Tissue culture of Pyrus hopeiensis was carried out using stem tips and bud tips of aseptic seedlings obtained from seed.Propagation coefficients in different hormone combinations with various concentrations were compared to investigate the effect of hormones on tissue culture reproductive capacity of P.hopeiensis.The results showed that"three lows"feature(low seed setting rate,low seed germination rate and low branch rooting rate)of P.hopeiensis is the main reason for its difficult population expansion,thus tissue culture using stems and buds of seedlings is an important way to solve the propagation problem of P.hopeiensis.The results also showed that the hormone ratio has significant influence on the tissue culture reproduction coefficient of P.hopeiensis,and 1/2MS medium with 6-BA(0.5 mg/L),NAA(0.05 mg/L)and TDZ(0.2 mg/L)was the best subculture medium.Using this subculture medium,the propagation coefficient could reach 3.91.We established a rapid propagation system of P.hopeiensis by tissue culture,and provided the theoretical basis and technical specifications for ex situ conservation of P.hopeiensis.
