卷叶贝母组织培养体系建立及ABA和GA3对其鳞茎萌发的影响

摘  要:本研究旨在探讨卷叶贝母组织培养体系建立,并外源添加激素脱落酸(abscisic acid,ABA),赤霉素(gibberellin acid,GA_(3))对其种子萌发特性的影响。本研究以卷叶贝母鳞茎为实验材料,在其培养基中添加不同激素及浓度,筛选卷叶贝母的最佳培养条件;在卷叶贝母鳞茎培养过程中,外源添加激素ABA和GA_(3),分析其萌发、营养物质及相关酶活性等生理指标变化。结果显示:(1)卷叶贝母组培培养最适条件:20℃,12 h/d的光照,诱导培养基为MS+0.5 mg/L 6-BA(6-苄氨基腺嘌呤)+2.0 mg/L NAA(萘乙酸),继代增殖培养基为MS+2.0 mg/L 6-BA+0.5 mg/L NAA。(2)鳞茎萌发特性:4℃冷藏期,GA_(3)可将正常40 d休眠期提前至29 d;4℃冷藏期,ABA处理可将正常休眠由40 d延长至50 d。(3)营养物质测定:休眠期间营养物质可溶性糖和可溶性蛋白含量呈上升趋势,而淀粉呈下降趋势。(4)相关酶活测定:解休眠期间相关酶活过氧化物酶(peroxidase,POD)和过氧化氢酶(catalase,CAT)呈下降趋势,而多酚氧化酶(polyphenol oxidase,PPO)呈上升趋势。总之,本研究成功地建立了卷叶贝母组织培养体系,并测定了外源激素ABA,GA_(3)对其鳞茎萌发特性的影响,为将来卷叶贝母的大规模繁殖提供支持。This research aims to investigate the establishment of a tissue culture system of Fritillaria cirrhosa,and the effects of exogenous addition of hormones such as abscisic acid(ABA)and gibberellin acid(GA_(3))on its seed germination characteristics.In this study,the bulbs of Fritillaria cirrhosa were used as experimental materials different hormones and concentrations were added to the culture medium to screen the best culture conditions of Fritillaria cirrhosa bulbs.During the cultivation of bulbs of Fritillaria cirrhosa,the hormones ABA and GA_(3) were added exogenously,then analyzed the changes of physiological indicators,such as germination,nutrients and related enzyme activities.Results:(1)Optimum conditions for tissue culture of Fritillaria cirrhosa:20℃,12 h/d light,and the induction medium is MS+0.5 mg/L 6-BA(6-benzylamino adenine)+2.0 mg/L NAA(Naphthalene acetic acid),the subculture medium is MS+2.0 mg/L 6-BA+0.5 mg/L NAA.(2)Bulb germination characteristics:GA_(3) can advance the normal dormancy period from 40 d to 29 d during the refrigeration period at 4℃;ABA treatment can prolong the normal dormancy period from 40 d to 50 d during the refrigeration period at 4℃.(3)Determination of nutrients:during the period of dormancy,the content of nutrient soluble sugar and soluble protein showed an upward trend,while starch showed a downward trend.(4)Determination of related enzyme activities:During the period of de-dormancy,the related enzyme activities Peroxidase(POD)and Catalase(CAT)showed a downward trend,while polyphenol oxidase(PPO)showed an upward trend.In a word,this study successfully established a tissue culture system of Fritillaria cirrhosa,and determined the effects of exogenous hormones ABA and GA_(3) on its bulb germination characteristics,and provided support for the large-scale reproduction of Fritillaria cirrhosa in the future.
