摘 要:目的 探讨低氧对细菌感染小鼠肠道固有免疫功能的影响。方法 将C57BL/6野生型小鼠随机分为常氧对照组(NC)、常氧感染组(NB)、低氧对照组(HC)和低氧感染组(HB)。以感染肠道鼠枸橼酸杆菌的小鼠为模型,分别在海拔2 260 m(大气压582mmHg, PO;121.6mmHg)和海拔5 000 m(大气压405mmHg, PO;84.7mmHg)处建立细菌感染模型,观察小鼠体重和结肠形态学变化;同时检测小鼠结肠、脾脏和血清中的中性粒细胞和巨噬细胞的相关细胞因子水平。结果 (1)HB组体重明显低于NB组(P<0.05);(2)HB组结肠组织有明显的黏膜破损和严重的炎细胞浸润,且HB组病理组织评分明显高于NB组(P<0.05);(3)HB组血清和结肠、脾脏中的中性粒细胞、巨噬细胞的炎性细胞因子及趋化因子明显低于NB组(P<0.05);(4)各感染组血清和结肠、脾脏中的中性粒细胞与巨噬细胞相关细胞因子及趋化因子水平均与相应对照组存在差异,差异有统计学意义(P<0.05)。结论 低氧会降低小鼠肠道中性粒细胞和巨噬细胞的趋化性,造成小鼠肠道固有免疫功能的降低;低氧可加重鼠枸橼酸杆菌感染导致的小鼠肠道病理变化。Objective To investigate the effects of hypoxia on the function of intestinal innate immunity in mice infected with bacteria.Methods C57 BL/6 wild mice were randomly divided into normoxia control group(NC),normoxia infection group(NB),hypoxia control group(HC)and hypoxia infection group(HB).The bacterial infection models were established at the altitude of 2 260(atmospheric pressure 582 mmHg, PO;121.6 mmHg)and 5 000 m(atmospheric pressure 405 mmHg, PO;84.7 mmHg)respectively.The mice were infected with Citrobacter rodentium as the experimental model.The variations of body weight and colon morphology were observed as well;the levels of centriocytes, macrophages and related cytokines in the colon, spleen, and serum of mice were also detected.Results(1)The body weight of HB group was significantly lower than that of NB group(P<0.05);(2)The colonic tissue of HB group had obvious mucosal damage and severe inflammatory cell infiltration.The pathological tissue score of HB group was significantly higher than that of NB group;(3)The inflammatory cytokines and chemokines related to neutrophils and macrophages in the serum, colon and spleen of HB group were significantly lower than those of NB group(P<0.05);(4)The levels of neutrophils, macrophage-related cytokines and chemokines in the serum, colon and spleen of each infection group were different from those of corresponding control group, and the difference was statistically significant(P<0.05).Conclusion Hypoxia can reduce the chemotaxis of neutrophils and macrophages in the intestine of mice, resulting in the decrease of intestinal innate immune function;the intestinal pathological variations caused by the infection of intestinal Citrobacter rodentium can be aggravated by hypoxia.
