摘  要:目的 考察丙二醇、氮酮、薄荷醇对黄香愈肤乳膏中盐酸小檗碱体外透皮吸收的影响。方法 采用Franz扩散池法,以20%乙醇-生理盐水溶液为接收液,以离体大鼠背部皮肤为模型进行体外透皮实验;采用HPLC法测定不同时间点接收液中盐酸小檗碱的含量,计算累积渗透量及稳态透皮速率,绘制累计渗透曲线;选择零级动力学模型、一级动力学模型和Higuchi方程模型对曲线进行拟合分析,测定取样末点盐酸小檗碱的皮肤滞留量。结果 以2%丙二醇作为促渗剂时黄香愈肤乳膏中盐酸小檗碱累计渗透量及稳态透皮速率均显著升高,黄香愈肤乳膏中的盐酸小檗碱体外透皮规律更符合一级动力学模型。结论 2%丙二醇对黄香愈肤乳膏中的盐酸小檗碱具有最佳促透效果。Objective To investigate the effects of propylene glycol, azone and menthol on the in vitro transdermal absorption of Berberine hydrochloride in Huangxiang healing cream.Methods Franz diffusion cell method was used to carry out in vitro transdermal experiment with 20% ethanol-physiological saline solution as receiving solution and isolated rat back skin as model.The content of berberine hydrochloride in the receiving solution at different time points was determined by HPLC.The cumulative penetration and steady-state transdermal rate were calculated, and the cumulative penetration curve was drawn.Zero-order kinetic model, first-order kinetic model and Higuchi equation model were selected to fit and analyze the curve.The skin retention of berberine hydrochloride at the end of sampling were determined.Results The cumulative penetration amount and steady-state penetration rate of berberine hydrochloride in the cream both increase significantly taking 2% propylene glycol as penetration enhancer.The in vitro transdermal penetration law of berberine hydrochloride in the cream was more in line with the first-order model.Conclusion 2% propylene glycol has the best penetration promoting effect on berberine hydrochloride in Huangxiang healing cream.
