摘 要:目的:分析1型糖尿病(type 1 diabetes mellitus,T1DM)患者外周血环状RNA (circular RNA,circRNA)差异表达情况,鉴定潜在分子标记物。方法:选取2021年7—12月于汕头大学医学院第一附属医院内分泌科住院治疗的1型糖尿病患者3例(T1DM组)和同期健康对照者3名(对照组)。采用circRNA表达谱芯片进行外周血样本检测。筛选两组差异谱,进行实时荧光定量PCR验证确定差异,并预测与circRNA结合的靶微RNA。结果:circRNA上调5 134个,下调5 426个,69个circ RNA有显著改变(P<0.05,|log FC|>1.5)。选取|log FC|改变更为明显的5个circ RNA,通过实时荧光定量PCR验证发现hsa_circ_101079、hsa_circ_100332、hsa_circ_085129及hsa_circ_103845表达与对照组相比差异均有统计学意义(P值均<0.05)。结论:hsa_circ_101079、hsa_circ_100332、hsa_circ_085129及hsa_circ_103845可能通过靶微RNA调控相关信号通路参与T1DM的发生发展。Objective: To investigate the different expression of peripheral blood circular RNA(circRNA) and identification of potential molecular markers in type 1 diabetes mellitus(T1DM). Methods: From July 2021 to December 2021, 3 patients with T1DM and 3 contemporaneous period healthy persons(control group) were selected from the Department of endocrine, the First Affiliated Hospital of Shantou University Medical College.Peripheral blood samples were detected by circRNA expression profiling chip. After screening the 2 different spectrum groups, real-time quantitative PCR(qPCR) was validated to confirm the difference. The target microRNA combined with circRNA were predicted. Results: 5 134 circRNAs were up-regulated and 5 426 were down regulated. There were significant changes in 69 circRNAs(P<0.05, |logFC|>1.5). Five circRNAs with more obvious |logFC| changes were selected and verified by q PCR. The expression of hsa_circ_101079, hsa_circ_100332, hsa_circ_085129 and hsa_circ_103845 were significantly different from that of the control group.Conclusion: hsa_circ_101079、hsa_circ_100332、hsa_circ_085129 and hsa_circ_103845 may be involved in the occurrence and development of T1DM through the regulation of related signal pathways by target microRNAs.
