摘  要:克隆猪三联基序21(Tripartite motif 21,TRIM21)基因,并对其进行序列分析和生物学功能预测。采用PCR技术扩增猪TRIM21基因,将其克隆至pCE2 TA/Blunt-Zero载体上。对猪TRIM21基因生物学功能进行预测,并对其理化性质进行分析。核苷酸相似性与进化树分析结果显示,猪TRIM21基因与黄牛核苷酸相似性为86.0%,亲缘关系最近,与斑马鱼核苷酸相似性为47.7%,亲缘关系最远;序列分析结果表明猪TRIM21基因序列全长1401 bp,编码469个氨基酸,编码蛋白的分子量为53.57 ku,理论等电点为6.07,属于亲水性蛋白。成功获得猪TRIM21基因序列,初步通过生物学信息学分析了猪TRIM21基因,为进一步揭示猪TRIM21基因的蛋白表达和免疫学特点提供理论依据。This study was to clone porcine Tripartite Motif 21(TRIM21)gene,and to analyze its sequence and predict its biological function.Porcine TRIM21 gene was amplified by PCR and cloned into pCE2 TA/Blunt Zero vector.The biological function of porcine TRIM21 gene was predicted and its physical and chemical properties were analyzed.Nucleotide similarity and evolutionary tree analysis showed that the nucleotide similarity of porcine TRIM21 gene with cattle was 86.0%,the nucleotide similarity with zebrafish was 47.7%,and the nucleotide similarity with zebrafish was the furthest.Sequence analysis showed that the total length of pig TRIM21 gene was 1401 bp,encoding 469 amino acids and molecular weight of 53.57 ku with a theoretical isoelectric point of 6.07.In this study,the porcine TRIM21 gene sequence was successfully obtained,and the biological information of porcine TRIM21 gene was preliminarily studied,providing theoretical basis for further revealing the protein expression and immunological characteristics of porcine TRIM21 gene.
