Effects of Rhynchophylline on Angiotensin,Aldosterone and Calcitonin Gene-related Peptide in Renal Hypertensive Rats

摘  要:目的探讨钩藤碱对肾性高血压大鼠血管紧张素、醛固酮(ALD)、降钙素基因相关肽(CGRP)的影响并分析其降压机制。方法将60只无特定病原体(SPF)级SD大鼠随机分为空白组、模型组、卡托普利组、钩藤碱高浓度组、钩藤碱中浓度组、钩藤碱低浓度组,每组10只。除空白组外,其余5组均建立肾性高血压模型。卡托普利组给予40 mg/kg卡托普利灌胃;钩藤碱高、中、低浓度组分别给予钩藤碱400 mg/kg、200 mg/kg、100 mg/kg灌胃;空白组、模型组给予等量生理盐水灌胃,每日1次,连续灌胃6周。在造模前、造模成功后4周、末次(6周)给药2 h后测量大鼠尾动脉压;处死大鼠,冲洗后天平称取左右肾重量;分离血浆,采用放射免疫法测定大鼠血浆ALD、血管紧张素Ⅰ(AngⅠ)、血管紧张素Ⅱ(AngⅡ)及CGRP含量;切除大鼠胸主动脉组织,离心匀浆后检测AngⅡ含量。结果与空白组比较,模型组造模后4周、给药6周后血压明显升高(P<0.05);造模后4周,模型组、卡托普利组、钩藤碱高浓度组、钩藤碱中浓度组、钩藤碱低浓度组血压均较造模前高(P<0.05);与造模后4周比较,给药6周后卡托普利组、钩藤碱高浓度组、钩藤碱中浓度组、钩藤碱低浓度组血压均较造模后4周高(P<0.05)。与空白组比较,模型组大鼠血浆AngⅠ、AngⅡ、ALD及CGRP含量明显升高(P<0.05);与模型组比较,卡托普利组、钩藤碱高浓度组、钩藤碱中浓度组、钩藤碱低浓度组大鼠血浆AngⅡ、ALD含量明显下降,AngⅠ、CGRP含量明显增高(P<0.05)。与空白组比较,模型组大鼠主动脉组织AngⅡ含量明显升高,与模型组比较,卡托普利组、钩藤碱高浓度组、钩藤碱中浓度组、钩藤碱低浓度组大鼠主动脉组织AngⅡ含量明显降低(P<0.05)。与空白组比较,模型组大鼠左肾质量、左右肾质量比值明显下降(P<0.05);与模型组比较,卡托普利组、钩藤碱高浓度组、钩藤碱中浓度组、钩藤�Objective To investigate the effects of rhynchophylline on angiotensin,aldosterone,and calcitonin gene-related peptide in renal hypertensive rats and to analyze its antihypertensive mechanism.Methods A total of 60 Sprague-Dawley(SD)rats without specific pathogen(SPF)were randomly divided into blank group,model group,captopril group,high concentration of rhynchophylline group,medium concentration of rhynchophylline group,and low concentration of rhynchophylline group,with 10 rats in each group.Except blank group,in the other 5 groups renal hypertension model were established.Captopril group was given 40 mg/kg captopril by gavage;high,medium,and low concentration of rhynchophylline groups were given 400 mg/kg,200 mg/kg,and 100 mg/kg of rhynchophylline,respectively;blank group and model group were given isometering normal saline by intragastric administration,once a day,for 6 weeks.Caudal artery pressure was measured before modeling,at 4 weeks after modeling,and 2 hour after the last administration;the rats were sacrificed and the left and right kidneys were weighed after washing;aldosterone(ALD),angiotensinⅠ(AngⅠ),angiotensinⅡ(AngⅡ),and calcitonin gene-related peptide(CGRP)in plasma were determined by radioimmunoassay;the content of AngⅡwas detected after the thoracic aorta tissue was resected and homogenized by centrifugation.Results Compared with the blank group,blood pressure in model group increased significantly at 4 weeks after modeling and at 6 weeks after administration(P<0.05);at 4 weeks after modeling,blood pressure of model group,captopril group,high concentration of rhynchophylline group,medium concentration of rhynchophylline group,and low concentration of rhynchophylline group were higher than those before modeling(P<0.05);compared with data at 4 weeks after modeling,blood pressure was higher in captopril group,high concentration of rhynchophylline group,medium concentration of rhynchophylline group,and low concentration of rhynchophylline group after 6 weeks administration(P<0.05).Compared
