摘 要:目的探讨当归芍药散对快速老化小鼠学习记忆的影响及作用机制。方法选取快速老化SAMR1小鼠10只、SAMP8小鼠30只,采用随机数字表法对实验动物进行编号,按照机遇均等原则分为SAMR1组、SAMP8组、DSS 15 g/kg组和DSS 60 g/kg组,每组10只。SAMR1组小鼠作为对照组,DSS 15 g/kg组和DSS 60 g/kg组小鼠分别给予15 g/kg、60 g/kg剂量的当归芍药散灌胃,每日1次;SAMR1组和SAMP8组小鼠给予相应体积的生理盐水(每10 g 0.1 mL)灌胃,每日1次。连续灌胃8周后,进行Y迷宫实验和Morris水迷宫实验检测小鼠的学习和记忆能力。行为学实验结束后,取出各组大鼠海马组织,采用酶联免疫吸附测定法(ELISA)检测组织裂解物中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)水平。结果小鼠Morris水迷宫行为学实验发现,在连续5 d寻找水下平台测试中,与SAMR1组小鼠比较,SAMP8组小鼠寻找水下平台的日均潜伏期明显延长,穿越平台象限时间和距离占整个游泳时间或距离的百分比下降(P<0.05);而DSS 60 g/kg组小鼠寻找水下平台的日均潜伏期较SAMP8组明显缩短,穿越平台象限时间和距离占整个游泳时间或距离的百分比均上升(P<0.05)。与SAMR1组小鼠比较,SAMP8组小鼠进臂正确率明显下降(P<0.05);与SAMP8组小鼠比较,DSS 60 g/kg组小鼠进臂正确率明显增加(P<0.05)。与SAMR1组小鼠比较,SAMP8组小鼠海马组织中TNF-α、IL-1β、IL-6的表达量明显升高(P<0.05);与SAMP8组小鼠比较,DSS 60 g/kg组小鼠海马组织中TNF-α、IL-1β、IL-6的表达量明显下降(P<0.05)。结论当归芍药散可能通过抑制SAMP8小鼠海马组织TNF-α、IL-1β、IL-6的表达来缓解SAMP8小鼠认知功能障碍。Objective To explore the effect and mechanism of Danggui Shaoyao San(DSS)on learning and memory of rapidly aging mice.Methods Rapidly-aging SAMR1 mice of 10 and SAMP8 mice of 30 were randomly divided into SAMR1 group,SAMP8 group,DSS15 g/kg group,and DSS60 g/kg group,with 10 mice in each group.SAMR1 mice were used as control group,mice in DSS15 g/kg group and DSS60 g/kg group were given 15 g/kg and 60 g/kg Danggui Shaoyao San gavage,respectively,once a day.Mice in SAMR1 group and SAMP8 group were given corresponding volume of normal saline(0.1 mL/10 g)by gavage,once a day.After 8 weeks of continuous intragastric administration,Y maze test and Morris water maze test were performed to detect the learning and memory ability of mice.After the behavioral experiment,the hippocampal tissue of each group was taken out,and the levels of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),and interleukin-6(IL-6)in tissue lysates were detected by enzyme-linked immunosorbent assay(ELISA).Results Morris water maze behavior experiment of mice showed that in the test of searching for underwater platform for 5 consecutive days,compared with SAMR1 group,the average latency of searching for underwater platform in SAMP8 group was significantly longer,and the percentage of time and distance across platform quadrant in the whole swimming time or distance was decreased(P<0.05);compared with SAMP8 group,the average latency of DSS 60 g/kg group searching for underwater platform was significantly shorter,and the percentage of crossing platform quadrant time and distance in the whole swimming time or distance were increased(P<0.05).Compared with SAMR1 group,the accuracy of arm entry in SAMP8 group were significantly decreased(P<0.05),and compared with SAMR8 group,the accuracy of arm entry in DSS 60 g/kg group was significantly increased(P<0.05).Compared with SAMR1 group,the expression levels of TNF-α,IL-1β,and IL-6 in hippocampus of SAMP8 group were significantly increased(P<0.05),and compared with SAMR8 group,the expression levels
