摘  要:目的 探讨miRNA标记物在骨髓间充质干细胞对成骨细胞诱导分化过程中的调控机制。方法 选取2016年6月—2018年1月该院收治的40例脊柱结核患者作为研究对象,将体外培养的人骨髓间充质干细胞(BMSCs)通过转化生长因子-β1(TGF-β1)对骨髓间充质干细胞向成骨细胞分化进行诱导,通过基因芯片技术对诱导分化期间miRNA进行检测,最后通过实时荧光定量PCR进行验证。结果 骨髓间充质干细胞向成骨细胞分化期间筛选出差异表达miRNA,其中miR-130b、miR-193b为表达上调miRNA,miR-135、miR-424为表达下调miRNA。于原样本中实施实时荧光定量PCR验证,验证结果同基因芯片筛选结果存在高度一致性(Kappa=0.913,P<0.05)。结论 高表达的miR-130b、miR-193b及低表达的miR-135、miR-424均有可能够调控骨髓间充质干细胞对成骨细胞的诱导分化过程。Objective To investigate the regulatory mechanism of miRNA markers in the induction and differentiation of bone marrow mesenchymal stem cells to osteoblasts.MethodsForty patients with spinal tuberculosis admitted to the hospital from June 2016 to January 2018 were selected as the study objects.Human bone marrow mesenchymal stem cells(BMSCs) cultured in vitro were induced to differentiate into osteoblasts by transforming growth factor-β1(TGF-β1).miRNAs were detected during induced differentiation by gene chip technology,and finally verified by real-time fluorescent quantitative PCR.ResultsDifferentially expressed miRNAs were screened during the differentiation of bone marrow mesenchymal stem cells into osteoblasts.Among them,miR-130b and miR-193b were upregulated miRNAs,miR-135 and miR-424 were down-regulated miRNAs.Real-time fluorescence quantitative PCR was performed in the original sample,and the verification results were highly consistent with the gene chip screening results(Kappa=0.913,P<0.05).ConclusionHigh expression of miR-130b and miR-193b and low expression of miR-135 and miR-424 may be able to regulate the induction and differentiation of bone marrow mesenchymal stem cells to osteoblasts.
