禽腺病毒强毒(FAdV-4)WZ株对SPF鸡和SPF鸭的致病性比较

摘　　要：对28日龄的SPF鸡和SPF鸭通过肌肉注射10;TCID;血清4型禽腺病毒(FAdV-4)WZ株,系统比较了SPF鸡和SPF鸭的临床症状、大体病变、存活率、体质量、组织病理学变化,并通过实时荧光定量PCR方法检测组织病毒载量和泄殖腔排毒情况。结果显示,FAdV-4强毒株可致SPF鸡发生严重的包涵体肝炎-心包积液综合征(HHS)且导致100%死亡,在多种组织中观察到组织病理学损伤,并检出FAdV-4,且在感染后1 d即可检测到排毒。相反,SPF鸭在攻毒后14 d的观察期内均未出现发病和死亡;与对照组SPF鸭相比体质量增加趋势一致,差异不显著(P>0.05);剖检未见病变和组织病理学损伤;在鸭内脏组织样品和泄殖腔拭子中均未检出FAdV-4。结果表明,FAdV-4强毒株对SPF鸡具有很强的致病性,而对SPF鸭并无致病性。本研究通过比较FAdV-4强毒对鸡和鸭易感性的差异,证实FAdV-4强毒只感染鸡不感染鸭。In order to compare the pathogenicity of virulent FAdV-4 strain to SPF chickens and SPF ducks,28-day-old SPF chickens and SPF ducks were injected intramuscularly with 10;TCID;FAdV-4 WZ strain,then the clinical symptoms,gross lesions,survival rate,body weight,histopathological changes of SPF chickens and SPF ducks were compared systematically,as well as the tissue viral load and cloacal detoxification were detected by fluorescent quantitative real-time PCR.It was found that the virulent FAdV-4 strain could induce severe HHS with 100%mortality in SPF chickens,histopathological lesions and FAdV-4 were detectedin a variety of tissues,while cloacal detoxification could be detected from the first day after infection.On the contrary,SPF ducks had no morbidity or death during the 14-day observation period after challenge,the trend of weight gain was consistent with the control SPF ducks,and the difference was not significant(P>0.05),no lesions and histopathological damage were seen at necropsy,FAdV-4 was not detected in visceral tissue samples and cloacal swabs of ducks.The results indicated that the virulent FAdV-4 strain had strong pathogenicity to SPF chickens,but not to SPF ducks.By comparing the differences in susceptibility of virulent FAdV-4 to chickens and ducks,this study confirmed that virulent FAdV-4 only infects chickens and does not infect ducks.
